Analytical chemistry is an area of chemistry primarily focused on the study and use of instruments for separation, identification, and quantification of an analyte of interest. Specifically, separation science within analytical chemistry often refers to the process of dividing mixtures into their small component parts based on differences in their physical and chemical properties; this can be preparative or analytical chromatography. With advances in microfabrication and microfluidic technologies in recent years, miniaturization of the analytical process has been a priority. However, traditional preparative scale separation processes are equally impactful because of their use for isolating components on a greater scale to allow for isolation of highly pure products for further use. These traditional preparative separation methods, when employed in natural product extraction and isolation, can yield a highly purified product in enough quantity to perform further studies. The first research chapter describes the use of centrifugal partition chromatography (CPC) for the preparative-scale isolation and purification of xylindein from the fungi, Chlorociboria aeruginosa. Solvent system compositions for xylindein isolation using CPC were explored, resulting in a new solvent system appropriate for purification of xylindein. The separation technique was optimized and used to isolate naturally sourced xylindein in amounts suitable for further study. Though preparative scale separations are useful and impactful for natural product extraction, interest in miniaturization and microfluidic technologies has grown tremendously in the past decades owing to their benefits in many fields of applications. In the following chapter, a novel approach to fabrication employing a high-speed CO2 laser for etching was used for rapid fabrication of polymethyl methacrylate-based microfluidic devices. The fabrication method provides a simple approach for the production of microfluidic chips, and offers the versatility needed to achieve different designs and features for a wide variety of applications. The microfluidic chips fabricated using this technique were used to perform on-chip electrophoresis with rhodamine B as a standard test dye. The electropherograms obtained from the fabricated microchips were reproducible and comparable to a polymethyl methacrylate (PMMA) standard chip. Microfluidic devices can integrate the entire analytical process, including sample handling, preparation, reaction, separation, and detection, onto a single chip. The system also allows for size reduction of instrumental components, and enhances the development of portable analytical tools for field-testing applications. The final section of this work includes the utilization of a smartphone as the detection platform for a well developed, relatively inexpensive, commercially available clinical chemistry assay as a model for rapid and inexpensive field-portable diagnostic testing. It was possible to quantify glucose over its clinically important concentration range (30–515 mg/dL) with good linearity (R2 = 0.9994, n = 5). Data collected using the iPhone 4 and canine serum samples were in agreement with results from the instrumental “gold standard” (Beckman Coulter AU480 Chemistry System). Presented herein are applications in analytical chemistry and separation science. Preparative scale isolation of naturally extracted xylindein was achieved in good yield using CPC; a novel fabrication technique for a polymeric microfluidic device was developed and used for on-chip CE; the applicability of smartphone technology for glucose detection was demonstrated for use as a portable analytical tool. Finally, though analytical chemistry is a broad field, the methods and techniques described within this dissertation have contributed to furthering science and knowledge in the subfields of preparative scale separations, microfabrication and microfluidic technology, and smartphones as detectors for clinically relevant analytes.

Along with increasing security measures in Android platforms, the amount of Android malware that use remote exploits has grown significantly. Using mobile botnets, attackers concentrate on reliable attack vectors such as SMS messages. Short Message Service (SMS) has been increasingly targeted by a number of malicious applications ("apps") that have the ability to abuse SMS features in order to send spam, to transfer command and control (C&C) instructions, to distribute malicious applications via URLs embedded in text messages, to send text messages to premium-rate numbers, and to exploit smartphones. Efficient detection and defence techniques that use filtering and blocking methods for SMS botnets is therefore an urgent necessity. Unfortunately, most botnet detection solutions proposed so far are reactive; that is, they require a large amount of data in order to effectively generate signatures and filtering rules to differentiate between normal and malicious SMS messages. By using proactive approaches such as a multi-agent system, agents can monitor certain environments and report abnormal behaviour in order to protect user data. In this thesis, we propose an SMS-based botnet detection framework using intelligent agents that are used to detect malicious SMS messages and monitor smartphone resources which are typically targeted by SMS botnet attacks. The proposed detection framework is based on a multi-layer model which consists of three modules and intelligent agents. The first is an SMS signature-based detection module which can be used to combat SMS botnets, in which we first apply pattern-matching detection approaches for incoming and outgoing SMS text messages, and then use rule-based techniques to label unknown SMS messages as suspicious or normal. The second module, an anomaly-based detection module, employs unsupervised learning techniques, using clustering algorithms to group SMS messages into four class labels and to classify reported text messages to one of those four classes. The module also uses a robust and efficient behavioural profiling analysis to detect whether there are any correlations between classification results and alerts from profiling analysis. Rule-based correlations are used to label SMS messages as either normal or malicious. The third module is a defence module that can be used as a more proactive approach which directly generates signatures and rules in order to protect Android smartphones from abuse by SMS botnets. This module is used to generate signatures of malicious SMS messages, to update phone number blacklists, to analyze malicious applications and to send feedback to Android smartphones so that the user can take action. Finally, a multi-agent system that can be used to observe Android mobile devices and to interact with service provider agents in order to detect malicious applications and SMS botnet activities on Android mobile devices. We have developed an intelligent and proactive framework that scans incoming and outgoing text messages, monitors Android resources and observes user usage that includes user connectivity time. The framework creates a user profile that is used to perform behavioural profiling analysis in order to identity malicious SMS and cut the C&C Channel. The proposed framework has been implemented using JADE agents. We demonstrate the capability of the multi-agent system, signature-based detection, anomaly-based detection module, and defence module in accurately detecting SMS botnets, we conduct different experiments in three phases. In the first phase, we focus on evaluating the efficiency of the SMS signature detection module in Android devices. This module was evaluated using over 12,000 test messages. It was able to detect all 747 malicious SMS messages in the dataset (100% detection rate with no false negatives). It also flagged 351 SMS messages as suspicious. A comprehensive performance analysis of the anomaly-based detection module is conducted in the second phase. The detection performance of the anomaly-based detection module has an average accuracy of 95% and an average of false negative rate is 3.95% on applied datasets. After having studied the performance of each module individually, in the last phase, we analyzed the overall performance of the proposed framework and provided a thorough analysis of JADE agents monitoring mechanism after demonstrating the capability of each module individually. We used approximately 60,000 test messages to evaluate the proposed framework. The signature detection agents reported 165 malicious SMS messages and 3,081 suspicious SMS messages. The anomaly-based detection module labelled 941 SMS messages as malicious.

In this dissertation, I present several novel optical devices for biomedical imaging and spectroscopy. They are all capable of promoting the performance of existing imaging and spectroscopy applications or enabling new applications.In Chapter two, I present a laser based light source which is ultrafast, multi-line, dual polarization, broadly tunable, and power scalable. This light source could be an ideal source for nonlinear optical imaging. We combine soliton self-frequency shift and pulse division method together to achieve these features. Specifically, power scalable is achieved through pulse division and recombination, which provides a general solution for conventional soliton self-frequency shift source whose pulse energy is limited. Also, benefited from the pulse division, divided pulses can be shifted individually, which enables multi-wavelength and dual polarization capability. Simultaneous dual-polarization second-harmonic generation imaging is demonstrated based on these features. In Chapter three, I present a biodegradable step-index optical fiber, which is suitable for light delivery under in vivo condition. The fiber has desirable mechanical and biological properties which are superior to traditional silica optical fiber. This is attributed to the programmable citrate-based material platform we use to fabricate the fiber. The fiber has a 0.4dB/cm loss, with a 1/e length longer than 10cm. By using this fiber, successful light delivery in tissue as well as fluorescence excitation and collection are accomplished, which prove the fibers usefulness for in vivo applications. Further, successful image delivery verifies the fibers ability for image transmission, which can potentially enable endoscopy applications. In Chapter four, I demonstrate two smartphone based detection devices, suitable for spectroscopy applications. Both of them benefit from the smartphone, which is ideal for point-of-care applications. Specifically, we developed a G-Fresnel smartphone based spectrometer. Successful protein concentration measurement using Bradford reagents demonstrates its ability for general spectroscopy applications. In additional to the smartphone spectrometer, we also developed a smartphone chloridometer based on a citrate chloride sensor. Through clinical verification, the device shows high accuracy and broad measurement range, which can potential serve as a device for cystic fibrosis diagnosis.

This SpringerBrief begins by introducing the concept of smartphone sensing and summarizing the main tasks of applying smartphone sensing in vehicles. Chapter 2 describes the vehicle dynamics sensing model that exploits the raw data of motion sensors (i.e., accelerometer and gyroscope) to give the dynamic of vehicles, including stopping, turning, changing lanes, driving on uneven road, etc. Chapter 3 detects the abnormal driving behaviors based on sensing vehicle dynamics. Specifically, this brief proposes a machine learning-based fine-grained abnormal driving behavior detection and identification system, D3, to perform real-time high-accurate abnormal driving behaviors monitoring using the built-in motion sensors in smartphones. As more vehicles taking part in the transportation system in recent years, driving or taking vehicles have become an inseparable part of our daily life. However, increasing vehicles on the roads bring more traffic issues including crashes and congestions, which make it necessary to sense vehicle dynamics and detect driving behaviors for drivers. For example, sensing lane information of vehicles in real time can be assisted with the navigators to avoid unnecessary detours, and acquiring instant vehicle speed is desirable to many important vehicular applications. Moreover, if the driving behaviors of drivers, like inattentive and drunk driver, can be detected and warned in time, a large part of traffic accidents can be prevented. However, for sensing vehicle dynamics and detecting driving behaviors, traditional approaches are grounded on the built-in infrastructure in vehicles such as infrared sensors and radars, or additional hardware like EEG devices and alcohol sensors, which involves high cost. The authors illustrate that smartphone sensing technology, which involves sensors embedded in smartphones (including the accelerometer, gyroscope, speaker, microphone, etc.), can be applied in sensing vehicle dynamics and driving behaviors. Chapter 4 exploits the feasibility to recognize abnormal driving events of drivers at early stage. Specifically, the authors develop an Early Recognition system, ER, which recognize inattentive driving events at an early stage and alert drivers timely leveraging built-in audio devices on smartphones. The rapid development of mobile devices over the past few years has opened up opportunities for smart health, bringing an inexpensive, convenient and efficient way for medical data collection and health monitoring. With various built-in sensors, advanced processors and wireless communication capabilities, mobile devices can be exploited to develop a broad range of healthcare applications. The specific goal of this dissertation is to design systems for achieving smart health with various mobile devices (e.g., smartwatch, smartphone and wireless mote). First, we design a smartwatch based system which leverages the built-in accelerometer to monitor the respiratory rate and body position during sleep. To calculate respiratory rate, we design a filter to extract the weak respiratory signal from the noisy accelerometer data collected on the wrist, and use frequency analysis to estimate respiratory rate from the data along each axis. Further, we design a multi-axis fusion approach which can adaptively adjust the estimates from the three axes and then significantly improve the estimation accuracy. To detect the body position, we apply machine learning techniques based on the features extracted from the accelerometer data. Second, we present another smartwatch based system which detects 8 common daily activities, including sitting, walking, running, going upstairs, going downstairs, eating, driving and sitting in a vehicle. By leveraging the built-in motion sensors on the smartwatch (i.e., accelerometer and gyroscope), a multi-level classification system is proposed which considers both detection accuracy and energy efficiency. Several practical issues are considered in developing the system to meet the specific requirements of the smartwatch, such as sensing capabilities and resource constraints of the smartwatch, and different wearing habits of smartwatch users. Third, we propose a smartphone based system to unobtrusively detect the sound-related respiratory symptoms occurred in a user's daily life, including sneeze, cough, sniffle and throat clearing. The built-in microphone on the smartphone is used to continuously monitor a user's acoustic data and uses multi-level processes to detect and classify the respiratory symptoms. Several practical issues are considered in developing the system, such as users' privacy concerns about their acoustic data, resource constraints of the smartphone and different contexts of the smartphone. Finally, we design targeted vaccination for infectious disease containment. To understand the disease propagation better, we collect student contact traces in a high school based on wireless motes carried by students. With our wireless system, we can record student contacts within the disease propagation distance, and then construct a disease propagation graph to model the infectious disease propagation. Based on this graph, we propose a metric called connectivity centrality to measure a node's importance during disease propagation and design centrality based algorithms for targeted vaccination. Further, we enhance our solution by exploiting the knowledge of the infected nodes which have been detected during vaccination.
