This publication comprises the proceedings of the Vaccine Cell Substrates 2004 conference. The purpose of this conference was to review current data and progress in the field of cell substrates, discuss the continued use of existing tests and the appropriateness of new ones at this time, and develop consensus recommendations on how to address these issues, by either recommending implementation or identifying research gaps that preclude decision-making. Scientific topics covered oncogenicity of cellular components, both latent viruses and cellular DNA, viral adventitious agent test methods, level of assurance provided by the current tests, bovine (and porcine) viruses in (primarily) bovine-derived raw materials, and bovine spongiform encephalopathy agents as potential contaminants of cell substrates. Also presented were tests that ensure the safety and quality of novel vaccine cell substrates for investigational vaccine production. Academia, regulators, the vaccine industry and the testing industry developing, regulating or testing viral vaccines will greatly benefit from the wealth of new findings.

Book Description :

Transboundary animal diseases (TADs) are a major threat to livestock. They are highly contagious or transmissible, and they have the potential to cause high morbidity and mortality in both susceptible animal populations and humans. In addition, not only are TADs detrimental to national economies, they are also a serious threat to world food security. This volume presents the proceedings of an international workshop on Vaccines and Diagnostics for Transboundary Animal Diseases that was held in Ames (Iowa, USA) in 2012. Experts and scientists from academia, industry and government reviewed the current status of vaccines and diagnostics for high priority TADs, decision-making and regulatory processes for veterinary biologics, and the roles and responsibilities of government agencies. The discussions also addressed achievements and gaps in vaccine and diagnostics development for 11 important TADs as well as the translation of research findings into licensed novel vaccines and diagnostics for high-priority TADs.

Book Description :

With the latest advances in analytical technologies, most biological products can now be extensively characterized in terms of their identity, heterogeneity and impurity profile. The currently available analytical methods (both physicochemical and biological) can characterize the primary, secondary and to some extent, the higher order structure of proteins. The sensitivity and selectivity of these methods allow for the identification and characterization not only of the desired protein component, but also many product-related substances and impurities as well as process-related impurities present in the drug preparations. Using an appropriate selection of analytical tools, it may be possible to demonstrate physicochemical and functional comparability between protein products manufactured before and after a manufacturing change or during product development. However, the extent of demonstrable comparability depends on the tests used, on the nature of the product and on the basic understanding of structure-function relationships. Assessment of immunogenicity is also a critical component of product characterization and is highly dependent on the types of assays used.

Book Description :

Fish farming, in seawater and in freshwater, in cages, tanks or ponds, makes an ever-increasing and significant contribution to the production of aquatic food in many regions of the world. During the last few decades there has been significant progress and expansion in the aquaculture sector, characterized by intensified production and the exploitation of many new species. Aquaculture must be a sustainable bio-production, environmentally as well as economically. Disease prevention in order to reduce losses, and the use of antimicrobials is crucial in this perspective. Vaccination has, in a few years, become the most important method for disease prevention in aquaculture, and effective prophylaxis based on stimulation of the immune system of the fish is essential for further development of the industry. This book provides general information about disease prevention in fish by vaccination, as well as specific descriptions of the correct use of vaccines against the most important bacterial and viral infectious diseases of aquatic animals. The book is written by some of the world’s leading experts in the subject, drawn from many countries where aquaculture is a significant and expanding part of the economy. Fish Vaccination is an encyclopedia of fish vaccinology for every present and future aquaculturist. Professionals in the aquaculture sector, including fish veterinarians and fish biologists, within the industry, in scientific institutions and regulatory authorities will all find a huge wealth of commercially important knowledge within this book. Libraries in all universities where aquaculture, biological and veterinary sciences are studied and taught should have copies of this important book on their shelves.

Book Description :

Improving the safety for blood and blood product transfusion It is amazing that in the first years of the 21st century there are insufficient blood supplies and that some supplies are unsafe in certain areas of the world. Regulations and codes call for continuous improvement in processes and attitudes to assure safety everywhere. To that end nearly 200 transfusionists and their colleagues gathered in Sydney in October 2005 to conduct a marathon symposium on methods and advances made towards achieving universal safety for blood and blood product transfusion. Sponsors of the latest products joined in to support this extraordinary meeting of information and ideas. Advances in transfusion safety were put forward from all areas of the blood transfusion process, including establishment of services, donor selection, infections that may affect donors, safe testing, risk and safety assessment, the roles of government and regulation, the use of blood and its products as well as the need for effective communication. The resulting papers from internationally renowned experts are published in these proceedings.

Book Description :

This volume brings together recent developments in the field of physico-chemical procedures and provides a perspective of the contribution that this methodology can make to facilitate the production and quality control of bacterial and viral vaccines. A growing number of vaccines today is based on well-defined materials, including vaccines based on purified protein antigens of natural origin or produced by rDNA technology, polysaccharides, semi-synthetic poly- or oligosaccharide-protein conjugates and novel nucleic acid constructs. Although in principle much better defined than 'traditional' vaccines, these new products raise new challenges of their own. The realization that biological assays often show limitations in characterizing these vaccines together with mounting social and political pressures to move away from test methods using live animals have shifted interest to methods providing a precise and objective means of characterization at the molecular level as an effective alternative. The growing sophistication of many procedures for physico-chemical analysis has made their application to biological materials a workable reality. This volume contains contributions describing the application of all the principal methods currently in use for evaluating carbohydrate, lipopolysaccharide, lipid, protein, glycoprotein, and lipoprotein components of vaccines, whether produced by conventional methods or recombinant technology. It is clear from the information presented that this approach is gaining momentum and that we are seeing only the beginning of its impact in the field of vaccines and vaccinology.

Book Description :

DNA vaccines have progressed rapidly from the conceptual stage to the stage of clinical trials. While studies in small laboratory animals have shown great promise, initial reports from human studies were less encouraging. Progress is being made, however, documented by the papers presented here. This volume contains the proceedings of a meeting devoted to the latest developments in DNA vaccines, from laboratory studies to clinical trials. The papers, written by leaders in the field, focus on the current state of DNA vaccines in humans and other large animals. The bulk of the studies involve DNA vaccines against HIV/SIV. Other promising trials make use of DNA vaccines against malaria and hepatitis B. The papers inform the reader about the immune basis of this form of vaccination and about approaches being developed to increase the efficacy of DNA vaccines in humans. These include the co-administration of cytokines, prime-boost strategies, optimising codon usage or the use of CpG motifs. An excellent up-to-the-minute account, this volume should be read by anyone in academia or in industry involved in DNA vaccination. The book will also be of great interest to clinicians involved in trials and those in the regulatory area.
