Raman scattering (RS) is a widely used vibrational technique providing highly specific molecular spectral patterns. This book is devoted to explanation of SERS in terms of the strong dipole and especially quadruple light-molecule interactions arising in surface fields strongly varying in space in the region of strongly irregular surface roughness. The influence of the quadruple interaction is a matter of principle, since it provides explanation for the appearance of forbidden bands arising in a great number of experiments on SERS on symmetrical molecules. This essential detail in fact is absent in all other SERS theories, that does not permit to create a closed theory, which explains the majority of experimental facts accompanying SERS by a unitary approach. Moreover as it is demonstrated in the book the huge enhancement in the phenomenon of Single Molecule Detection by the SERS method can be explained exclusively by the strong quadruple light-molecule interaction. The first part of the book is devoted to a brief description of main SERS characteristics and critical analysis of theoretical approaches. Further the theory of electromagnetic field near some model kinds of rough surfaces and some other systems and the theory of the SER cross-section for arbitrary and symmetrical molecules are presented in detail. The obtained expressions helps to establish selection rules for contributions in the SER cross-section and analyse the SER spectra of symmetrical molecules. This analysis corroborates main specific features of the SER spectra of symmetrical molecules and some anomalies, which exist in these spectra for some specific conditions. The existence of electrodynamic forbiddance of the quadruple scattering mechanism for the methane molecule and molecules with cubic symmetry groups is established. The above theory permits to explain not only the huge enhancement in the phenomenon of Single -Molecule Detection but the blinking of the SERS signal too. The first layer effect, arising in the first layer of absorbed molecules also is considered in detail. It appears that the nature of this phenomenon is the electrodynamic enhancement, which is very large just in the first layer. The other phenomena, accompanying SERS are accounted for. It is demonstrated that the theory is in a good agreement with the SERS experiments and explains quite a number of characteristics related to the SERS phenomenon.

Surface-enhanced Raman scattering (SERS) has long been the interest of researchers in chemistry, physics and engineering, especially since the discovery that SERS can probe into the system down to the single molecule (SM) level. Despite the large number of publications regarding the fabrication of SERS substrates, it has been a challenge in the field to quantify the SERS signal and universally compare substrates. Traditionally, enhancement factor (EF) is used as an indicator of substrate quality, but the EF calculation is hugely dependent on the estimation of the surface coverage and other factors that are determined largely subjectively. Therefore, this thesis aims at discussing other parameters that can also be used to evaluate different substrates. Six different SERS substrates of Ag or Au nanoparticles of different sizes were fabricated by nanosphere lithography (NSL) and characterized by electron microscopy and UV-vis spectroscopy. SERS substrates were mapped for different concentrations of a probe molecule. Through subsequent baseline correction and principle component analysis (PCA), the "intensity" of individual spectrum was obtained and the shapes of intensity histograms of each substrate were acquired.Instead of calculating EF, five criteria (six quantification methods in total) were employed to comprehensively evaluate the six substrates. These were density of hot spots (characterized by the number of zero-intensity events), enhancement (represented by mean intensity), spatial variation (calculated by RSD of intensity), repeatability (realized by cross correlation) and histogram shape (quantified by skewness and kurtosis). These new methods provide insights to the understanding of the properties of SERS substrates in terms of hot spots. Different substrates may exhibit better performance in terms of one criterion but worse in terms of others. Those variations in performance can be explained by their surface morphology.These more elaborated methods are believed to provide a more comprehensive approach to evaluate and compare substrates than the traditional EF values. The thesis also paves the way for future study on SM-SERS and fabricating better SERS substrates.

Cyanide is a toxin with many natural and industrial sources. Among several cyanide antidotes dimethyl trisulfide (DMTS) is one of the current promising candidates under investigation. Surface Enhanced Raman Spectroscopy (SERS) is a sensitive, cost effective method that has long term potential for multiplexed, portable sensing. In this research, experiments were carried out to compare three different approaches to sampling DMTS from ethanolic solutions: drop-coating, immersion, and headspace sampling. The SERS experiments in all media show promising sensitivity with detectable signals at 5 micromolar concentrations of DMTS in ethanol. Problems in the reproducibility of signals in the headspace sampling led to the observation that condensation on the sample cell windows and sensor was a problem. A proof of principle experiment using resistive heating to warm the sampling window was successful in removing and preventing condensation. Experiments were also carried out in which DMTS was reduced to produce more volatile products that could be detected more rapidly from the headspace than DMTS. In these experiments, SERS peaks were observed to grow in more rapidly than had been observed from headspace sampling above ethanolic DMTS solutions. Specifically, SERS peaks from reduction products were observed within a minute following the initiation of reaction.
