Genome-wide association studies (GWAS) for complex disorders with large case-control populations have been performed on hundreds of traits in more than 1200 published studies (http://www.genome.gov/gwastudies/) but the variants detected by GWAS account for little of the heritability of these traits, leading to an increasing interest in using family based designs. While GWAS studies are designed to find common variants with low to moderate attributable risks, family based studies are expected to find rare variants with high attributable risk. Because family-based designs can better control both genetic and environmental background, this study design is robust to heterogeneity and population stratification. Moreover, in family-based analysis, the background genetic variation can be modeled to control the residual variance which could increase the power to identify disease associated rare variants. Analysis of families can also help us gain knowledge about disease transmission and inheritance patterns. Although a family-based design has the advantage of being robust to false positives, novel and powerful methods to analyze families in genetic epidemiology continue to be needed, especially for the interaction between genetic and environmental factors associated with disease. Moreover, with the rapid development of sequencing technology, advances in approaches to the design and analysis of sequencing data in families are also greatly needed. The 11 articles in this book all introduce new methodology and, using family data, substantial new findings are presented in the areas of infectious diseases, diabetes, eye traits, autism spectrum disorder and prostate cancer.

The papers in this volume were presented as part of the University of Kansas Department of Anthropology Distinguished Lecture Program on Anthro pological Genetics. Consecutively, each contributor spent approximately a week on the campus at Lawrence participating in a seminar. The contributors to this volume were not on campus at one time, but visited us on alternating weeks; hence, a symposium-type interchange was not possible between all participants. However, the students and faculty of Kansas University acted as a sounding board. This volume can be considered a companion and continuation of Methods and Theories of Anthropological Genetics, which was based upon a symposium on the state of the art in 1971. This present volume reflects what we consider to be some of the advances and current developments in anthropological genetics since 1973. Emphasis has shifted, to some degree, away from population struc ture analysis (as depicted in Crawford and Workman) to genetic epidemiology. However, population structure still remains a fertile and ongoing area of research with many theoretical questions still remaining unanswered.

Prior and subsequentto the publication of the third edition of the Diagnos tic and Statistical Manual of Mental Disorders (DSM-III), we have witnessed a considerable upsurge in the quantity and quality of research concerned with the psychiatric diagnostic process. There are several factors that have contributed to this empirical influx, including improved diagnostic cri teria for many psychiatric disorders, increased nosological attention to childhood psychopathology, and development and standardization of several structured diagnostic interview schedules for both adult and child populations. With the advent of DSM-III-R, and in anticipation of DSM-IV, diagnostic labels and their definitions have been in a state of change, as evinced by the many refinements and modifications currently taking place. However, the basic purpose or raison d'etre of the nosological scheme has not been altered. Psychiatric diagnosis is the means by which we classify or categorize human psychopathology. And, as is the case in the medical arena, psychiatric diagnosis serves three central functions: classification. communication. and prediction. As research accumulates, our understanding of psychiatric disorders increases, and we are in a much better position to classify reliably and with validity, as well as to com municate and predict Despite periodic changes in the diagnostic system, the basic strategies for conducting diagnostic research (e. g. , genetic-family studies, biological markers, follow-up studies, etc. ) do not vary appreciably over time. But in over one decade no scholarly book has appeared that tackles the essential research issues involved in upgrading the diagnostic endeavor.
