An introduction to microfluidics with an emphasis on paper and thread utilization for microfluidic point-of-care (POC) diagnostic devices is described in Chapter 1. Colorimetric analysis for analyzing POC microfluidic devices (MDs) is discussed. Chapter 2 describes a novel microfluidic thread/paper-based analytical device (℗æTPAD) and microfluidic paper-based analytical device (℗æPAD) with an enzymatic encapsulation polymer to detect and asses glucose and acetylcholinesterase (AChE). The ℗æTPAD was fabricated using a single strand of nylon thread, hole-punched chromatography circles, enzymatic polyacrylamide (PA) sphere, single sided tape, and a platform for stability. The device yielded a correlation between yellow-brown intensity and glucose concentrations or yellow intensity and ATC concentration. Sample solutions with unknown concentrations of glucose in artificial urine were tested and results were compared to the calibration curve. Chapter 3 is the fabrication of novel 3D-℗æPADs to detect glucose and bovine serum albumin (BSA) through colorimetric assays is described. The 3D-ÎơPAD consisted of three (wax, heat pressed wax-printed paper, single-sided tape) layers. The device yielding a correlation between either yellow intensity and glucose concentration or cyan intensity and BSA concentration. Chapter 4 discussed two 3D-ÎơPADs for vertical capillary flow bioassays. Both platforms utilized Inkscape to generate a 3D-℗æPADs wax printed design on chromatography paper. A glucose, AChE, and BSA assay were tested on the devices.
