The cyclic purine nucleotides 3’,5’-cAMP and 3’,5’-cGMP are well-established second messengers. cGMP has recently been covered in a volume of the Handbook of Experimental Pharmacology (volume 191). In addition to 3’,5’-cAMP and 3’,5’-cGMP, so-called non-canonical cyclic nucleotides exist. These comprise the cyclic pyrimidine nucleotides 3’,5’-cCMP and 3’,5’-cUMP, the purine nucleotide 3’,5’-cIMP, the 2’,3’-nucleoside monophosphates and cyclic dinucleotides. In this volume of the Handbook of Pharmacology, word-leading experts in the field summarize our current knowledge on these non-canonical cyclic nucleotides, discuss open questions, future research directions and the pharmacotherapeutic implications. Special emphasis will be given to the emerging roles of 3’,5’-cCMP and 3’,5’-cUMP as second messengers with regard to generators, effectors, biological functions, inactivation and bacterial toxins. For some of the studies a fluorescent biosensor expressed in the cytosol, or targeted to mitochondria or the nucleus, was used in order to monitor autophagic degradation of the respective compartment/organelle. Here the assay was based upon a genetically encoded dual colour-emission biosensor. Rosella, the fusion of a relatively pH-insensitive fast-maturing variant of the red fluorescent protein DsRed, with a pH-sensitive variant of GFP. By virtue of being targeted to different compartments/organelles the dual colour nature of Rosella biosensor, allows unambiguous distinction between material delivered to the vacuole (red fluorescence, but pH-based loss of green fluorescence) and that localised in cytosol, mitochondria and the nucleus (red and green fluorescence retained). This novel assay allows not only fast and simple qualitative analysis, but also quantitative analysis of cells in a population undergoing autophagy. The autophagic degradation of the cytosol was found to be fully dependent on the macroautophagic machinery. On the other hand, autophagic degradation of the nucleus can be observed to occur 24 hours following nitrogen starvation and either independently of some ATG genes (e.g. ATG5 and ATG12), but does rely on others (e.g. ATG3 and ATG4). Other data concerning dependence on components of the NV-junction and complexes previously identified as being involved in microautophagic processes suggest that the process of nucleophagy observed and described in this thesis is different from the PMN.

Fungi are found in virtually every environment, and comprise a significant portion of the normal microflora of healthy individuals. Some species of fungi are aeroallergen sources capable of inducing sensitization and causing exacerbation of asthma and respiratory allergy. Others are transmissible between hosts and may cause no symptoms in healthy individuals. However, immune suppressed individuals may develop invasive disease marked by tissue invasion with a potential for widespread dissemination. Existing therapies for patients consist of antifungal drugs, yet these require prolonged administration with the possibility of adverse side effects, and may be rendered ineffective by the emergence of antifungal-resistant strains. It is therefore of interest to increase our understanding of host-pathogen interactions in order to facilitate the development of new therapies for individuals suffering from fungal infection and disease. These early interactions are shaped by an array of constituent and secreted factors that stimulate or inhibit host immune responses toward protective or detrimental immunity. Likewise, an array of preformed factors and tissue-resident cells provide early protection from fungal infection and provide extracellular signals that result in localized recruitment of inflammatory cells and determine the character of subsequent adaptive antifungal immunity. Together with the microfilament, microtubule and intermediate-filament networks, septins constitute an integral part of the eukaryotic cytoskeleton. Historically identified as proteins critical for septum formation in the budding yeast Saccharomyces cerevisiae, septin family GTPases are expressed and participate in the process of cytokinesis in most eukaryotes except higher plants. More than a dozen septin genes in mammals, together with various splice variants displaying tissue-specific expression patterns and flexible hetero-polymeric higher-order assembly achieve an unfathomable complexity superior to the other cytoskeletal components. Even though the initial studies in the septin field was restricted to their evolutionarily conserved role in cell division, strong expression of septins in the non-dividing cells of the brain generated great interest in understanding their role in neuronal morphogenesis and other aspects of cellular function. On one hand, recent developments indicate complex non-canonical roles for septins in diverse processes ranging from neuronal development to immune response and calcium signaling. On the other hand several lines of data including those from knockout models question the universal role for septins in animal cell cytokinesis. Mammalian hematopoietic cells seem to proliferate and efficiently undergo cytokinesis in the absence of pivotal septin proteins in a context-dependent manner. The lack of septin-dependence of hematopoiesis also opens the possibility of safely targeting septin-dependent cytokinesis for solid-tumor therapy. Thus the septin field is perfectly poised with novel roles for septins being discovered and the basic understanding on septin assembly and its canonical functions constantly revisited. The objective of this research topic was to provide an exclusive platform for discussing these rapid advances in the septin field.
