Abstract: This dissertation describes my Ph. D. work that focused on the synthesis and evaluation of novel nitric oxide donors, and the synthesis of derivatives of 5-fluorouracil. In chapter 1, a series of alkyl and aryl N-hydroxyguanidines were synthesized and demonstrated to act as substrates of nitric oxide synthases (NOS). The discovery of these non-amino acid hydroxyguanidines as novel substrates also led to the discovery of a novel-binding mode of NOS. In chapter 2, in order to achieve site specific delivery of nitric oxide (NO), a new class of glycosidase activated NO donors has been developed. Glucose and galactose were covalently coupled to 3-morphorlinosydnonimine (SIN-1), a mesoionic heterocyclic NO donor, via a carbamate linkage at the anomeric position. The beta-glycosides were successfully prepared for these conjugates, while the alfa glycosidic compounds were very unstable. The new stable sugar-NO conjugates could release NO in the presence of glycosidases. Such NO prodrugs may be used as enzyme activated NO donors in biomedical research. In chapter 3, a new sialated diazeniumdiolate has been synthesized and the glycosylation product was exclusively an alfa anomer. This new nitric oxide donor exhibited significantly improved stability as compared to its parent diazeniumdiolate salts and it could be efficiently hydrolyzed by neuraminidase to release nitric oxide with a Km of 0.14 mM. The sialic acid-NO conjugate would be a valuable prodrug that targets NO to influenza viruses. In chapter 4, two classes of prodrugs of 5-fluorouracil (FU) were synthesized. The first type is FU-NO conjugate. The synthesized compounds showed greater cytotoxicities than 5-fluorouracil for DU 145 human prostate and HeLa cancer cells. The second type is carbohydrate-FU conjugate. A new reverse glycosadition was also developed to prepare sugar conjugates. In chapter 5, 7-carboxylindazole, as well as 5- and 6-carboxylindazoles was synthesized. These compounds were designed as isosteres of 7-nitroindazole, a well-known inhibitor of NOS. However, they did not show potent inhibition to NOS.

Nitric oxide (NO) may modulate mitochondrial O2 consumption in the heart, and provide another level of respiratory control under normal or hypoxic conditions. NO and NO donors have also been proposed to have both beneficial and deleterious effects during ischaemia-reperfusion (IR) injury, and both of these maybe mediated by interactions of NO with mitochondria. The aims of the research in this thesis were to investigate (i) whether there is a specific form of mitochondrial nitric oxide synthase (NOS), (ii) which NO donors release NO spontaneously and which require bioactivation, (iii) whether different NO donors are protective or deleterious during IR injury, (iv) the effect of NO donors on mitochondrial membrane permeability transition pore (mPTP) opening. In this study, using 3 different techniques (Western blotting, immunoprecipitation and DAF fluorescence), no evidence was found to suggest the existence mitochondrial NOS in highly purified heart and liver mitochondria. However, it was possible to detect all forms of NOS in crude heart and liver mitochondria, and the same applied for the ryanodine receptor and caveolin, in "mitochondrial" fractions. Despite the fact that NO donors are commonly used in medicine and in research, in many cases it is still unclear as to how NO is released from the NO donors; whether it is spontaneously released or whether bioactivation is required. Using the NO- sensitive fluorescent dyes DAF-2 and DAF-2 DA, the results of this study showed that the NO donors DEA and SIN-l spontaneously released NO. SNAP, a donor that has been assumed to be spontaneous, showed no release of NO. Sodium nitroprusside did show significant spontaneous release of NO, but only at 5 and 10 mM. The novel NO donor NCX2057 was the only NO donor that showed release of NO by mitochondrial metabolism. The nitrates BDMN and ISMN both released NO in post-mitochondrial supernatant, indicating the involvement of cytosolic enzymes in NO donor metabolism. Previous studies have shown that NO can have both protective and deleterious effects in whole heart IR injury. In this study it was shown that the administration of the NO donors, SNAP and DEA before the onset of global ischaemia and throughout reperfusion, resulted in a concentration-dependent protection against IR injury. Beyond a concentration threshold level (40 ~ SNAP and 2 ~ DEA), the protection was lost. Since IR injury is critically regulated by the opening and closing of the mPTP during reperfusion, the effects of DEA on mPTP opening, mitochondrial Ca2+ uptake and membrane potential were investigated using isolated mitochondria. The lower concentrations of DEA (2-25 flM) caused partial mitochondrial membrane depolarisation, sensitised the mPTP to Ca2+ and promoted mPTP opening. By contrast, the higher concentrations of DEA depolarised the mitochondrial membrane potential, prevented Ca2+ uptake and inhibited mPTP opening. This biphasic effect observed could be attributed to the inhibition of cytochrome c oxidase, as potassium cyanide had similar effects on membrane potential, Ca2+ uptake and mPTP opening as DEA. DEA also inhibited mPTP opening under de-energised conditions, indicating that in addition to modulating mPTP opening via inhibition of cytochrome c oxidase, DEA can also directly interact and modify mPTP components, possibly via nitration.

"The medicinal benefits of the nitric oxide (NO) donor compounds termed diazeniumdiolates are well established and include antithrombic, cytostatic, and vasorelaxant activities. Additionally, fendiline, a coronary vasodilator, increases intracellular NO concentrations via interaction with the Ca2 cascade. Modification of fendiline with NO generates a new, lipophilic member of the diazeniumdiolate family, fendiline diazeniumdiolate (FDL-NONa), which has increased potential as a pharmaceutical NO donor and multiple applications in the coronary health market. This bifunctional vasodilator, when dissolved in methanol and buffer solution, releases NO to generate a half-life of 1.6 hours at 37°C and pH 7.4. Extension of this half-life can be achieved by varying the water insoluble particulate size of the donor in PBS. Furthermore, excellent binding of FDL-NONa to both glass and metal surfaces provides a direct correlation between coating thickness and NO release while decomposition of the compound in vivo generates species which pose no health concern. In additional [sic] to numerous functions within the cardiovascular system, NO has also been established as an indispensable participant in the wound healing cascade. Based on these findings, multiple NO-based wound dressings have been developed for employment in impaired wound healing scenarios. One example is the ascorbic acid/nitrate patch, which has displayed very promising results in both diabetic and lechmaniasis wounds, however has several drawbacks including donor half-life, patch stability and product manufacturing costs. To correct these problems, a new two-layer, PAN-NO (polyacrylonitrile modified with nitric oxide) pouch has been developed. Modification of previously electrospun PAN nanofibers results in the formation of a new polymeric PAN-NO patch that provides a 30 min increase in patch half-life, with extension of the total release time by 4 to 5 h and a nearly 17x increase in NO release from the four-layer, ascorbic acid/nitrite patch when modified in sodium trimethylsilanolate/1,4-dioxane or a 4 h increase in patch half-life, with an extension of the total release time by 27 to 28 h and a comparable NO/mg release when modified in sodium methoxide. Insertion of either form of PAN-NO into a heat-sealed, ascorbic acid poudh forms a second generation, NO-donating wound dressing, with reduced manufacturing costs and improved NO release kinetics."--Abstract.
