In addition to intact protein separation, a micellar liquid chromatography (MLC) system was developed using the polyacrylonitrile UTLC device for the size characterization of polyethylene glycol (PEG)ylated gold nanoparticle (AuNP), an emerging agent in cancer therapeutics, of which the cellular uptakes and cytotoxicity are highly dependent on its size. PEGylated AuNPs with different sizes in the range of 10-80 nm were well separated from each other. The developed method also permitted the separation of AuNPs capped with different molecular weight of PEG in the range of 2-20 kDa. Micellar mobile phases were adopted to provide a highly biodegradable chromatographic system. This method exhibited excellent separation performance with smallest plate heights 2 μm and resolution of each pair of AuNPs 1.5. Decent separations for all PEGylated AuNPs could be achieved within 5 min. This method was applied to monitor the transformation of AuNPs in serum protein, serving as a rapid and convenient tool for characterization of size distribution and modification of PEGylated AuNPs.

Tian Lu's dissertation describes major advances in ultrathin-layer chromatography (UTLC), liquid chromatography and surface-assisted laser desorption ionization (SALDI), and matrix-enhanced SALDI (ME-SALDI) mass spectrometry. Lu describes the fabrication of electrospun polyvinyl alcohol (PVA) UTLC plates using an in-situ crosslinking electrospinning technique. The author improved the efficiency of PVA plates greatly compared to the efficiency of silica HPTLC plates. Also highlighted in this thesis is an edge-plane based ordered-carbon surface that provides unique selectivity in liquid chromatography. Further developments include polar analytes, such as amino acids, nucleotides and nucleosides which can be well-retained and separated in the edge-plane ordered-carbon stationary phase. Also, the author studied and detected mass spectra of organic polymers as high as 900,000 Da, the highest molecular weight that has been studied by SALDI to date using the carbon nanofibrous substrate. This thesis has led to a number of publications in high-impact journals.

Abstract: Separation efficiency is inversely related to stationary phase diameter. Improvements in stationary phase manufacturing have led to micron-sized particles for packing. If packing materials could be decreased down to the nanometer scale, the band dispersion would theoretically be lowered by 6 orders of magnitude. However, the quest towards the use of nanomaterials in chromatography has encountered serious challenges such as extremely high back pressures and problems associated with frits. Ideal chromatographic methods to examine the performance of nano-stationary phases would be free of complicated packing methods. Work described in this dissertation illustrates two such techniques: ultra-thin layer chromatography (UTLC) and electrokinetic chromatography (EKC). Electrospinning is a simple and inexpensive method to create nanomaterials of different morphology (i.e. fibers, ribbons, beads) and was found to be an ideal way to fabricate TLC devices with nanoscale stationary phases. Polymer stationary phases such as polyacrylonitrile (PAN) as well as glassy carbon nanofibers demonstrate superior chromatographic properties in the separation of laser dyes, steroidal compounds and fluorescently labeled essential amino acids. Electrokinetic chromatography (EKC) has also been shown to be a nano-compatible chromatographic method. EKC takes advantage of a mobile, pseudo-stationary phase (PSP) plug rather than a packed or immobilized phase. Studies of ordered carbon nanomaterials and their retention characteristics are described using electrokinetic chromatography. Small organic molecules and their acids were used as initial analytes to elucidate the selectivity of the edge-plane carbon nanorods and compared to results obtained using amorphous carbon nanorods. A separation of 4 beta blocker pharmaceuticals were also separated using edge-plane carbon PSP. The electrical properties of unique carbon nanomaterials studied in this dissertation are also examined in Chapter 5 to aid in the understanding of these materials in an electrically driven method such as EKC.

Nanomaterials are attracting an interest of many researches. All this attention is due the unique physical and chemical properties of nanomaterials differing significantly from the bulk materials mainly due to their size in range of nanometers. Capillary electrophoresis (CE) is a powerful, well-established analytical technique that provides numerous valuable benefits over other separation methods including high-performance liquid chromatography. The connection between CE and nanotechnology can be approached by two strategies: (i) CE analysis of nanomaterials and (ii) nanomaterials for CE improvement. The first perspective focuses on uses of CE as a method for characterization employed during nanomaterial production and modification as well as for monitoring their properties and interactions with other molecules. The second viewpoint deals with applications of nanomaterials for improving CE performance, mainly by enhancing efficiency of separation using nanomaterials as a stationary or pseudo-stationary phase and by enhancing detection sensitivity and/or selectivity in both optical and electrochemical detection. Moreover, applications of nanomaterials for sample preparation before CE analysis will be mentioned. This chapter aims at highlighting the symbiosis of CE and nanotechnology as a combination of modern, progressive field with well-known and reliable analytical method.
