A critical first step in the design of mucosal vaccines is the selection of administration route. Not all mucosal immunization routes are created equally when it comes to eliciting immune responses in multiple body compartments. This subject and situations when a mucosal route may not be required for vaccine delivery are reviewed here with an emphasis on the sublingual immunization route, which may offer a safer alternative to the nasal route for induction of broadly disseminated immune responses. External host defenses that inhibit entry of microorganisms at mucosal surfaces also pose obstacles to the efficient internalization of mucosally-applied vaccines. Transcutaneous immunization with appropriate adjuvants and permeation enhancers can induce mucosal immune responses and may be advantageous for bypassing these luminal barriers. Other chapters describe strategies for enhancing uptake of mucosal vaccines, for instance through targeted delivery to antigen-sampling M cells, construction of virus-like particles which mimic natural pathogens, addition of mucoadhesives or formulation as nanoparticles. Topics include edible vaccines as well as plant-based production of subunit or particulate vaccines that could be administered by any route. Dry powder vaccines that could be insufflated or directly applied to mucosal surfaces may be particularly ideal for mass vaccination in developing countries. The manufacture, stability and efficacy of powder formulations is comprehensively reviewed. We conclude with chapters on two of the greatest challenges facing mucosal vaccine development: human immunodeficiency virus and bioterrorist agents. This monograph highlights progress and information that should prove invaluable for the development of contemporary vaccines that prevent infection by these and other mucosal pathogens.

Novel mucosal vaccines (LL-M2e, LL-HAe, SG-HAe) were constructed from live, non-pathogenic Lactococcus lactis or Streptococcus gordonii that express conserved regions of HA or M2 antigens from avian influenza virus (AIV) A. All three vaccines evoked antigen-specific serum immunoglobulin G (IgG) responses in vaccinated chickens. The addition of the adjuvant cholera toxin B (CTB) increased the antigen specific IgG response. Fecal IgA and cell mediated immune responses were not detectable in the chickens. LL-M2e provided significant protection against lethal challenge compared to mock-vaccinated chickens. None of the chickens vaccinated with the control strain (LL-PIP) or mock-vaccine survived lethal challenge past 5 days, whereas 2 of the 12 chickens vaccinated with LL-M2e survived at least 7 days post-infection. An M2e-specific serum IgG concentration of 1100 ng/ml was found to be the minimum required for increased protection against AIV infection. SG-HAe, but not LL-HAe, provided a significant increase in the number of chickens that survived lethal challenge compared to control-vaccinated birds. However, no correlation between HAe-specific serum IgG and survival was found.

The transition period is critical for the health and productivity of dairy cows. Feeding high-grain diets immediately after parturition is necessary to support high milk production. However, high-grain diets are associated with acidic rumen pH and alterations in the composition of rumen microbiota in favor of pathogenic Gram-negative and Gram-positive bacteria which releases considerable amounts of lipopolysaccharide (LPS) and lipoteichoic acid (LTA) in the rumen fluid. There is evidence for translocation of LPS into the host systemic circulation causing multiple metabolic and immune disturbances; however, no reports related to the fate of LTA in the rumen have been reported. Both LPS and LTA have been implicated in the etiopathology of multiple diseases in dairy cows and other animals. However, no strategies on how to prevent their deleterious effects have been explored so far in dairy cows. We hypothesized that mucosal (i.e., oral or nasal) vaccination against LPS and LTA before parturition, would stimulate mucosal immunity and confer protection to dairy cows. The overall aim was to enhance the immune competence and lower the occurrence of most frequent periparturient diseases after parturition. To reach this goal two studies were conducted. In study 1, 100 dairy cows were vaccinated several times oronasally either with saline (controls) or with increasing doses of LPS from Escherichia coli 0111:B4 before parturition. Thirty cows out of 100 were selected for intensive sampling. Results from this study indicated that oronasal treatment with LPS modulated plasma metabolic and immune profiles, increased total salivary and vaginal mucus immunoglobulin-A (IgA), lowered the incidence of selected periparturient diseases, and improved overall productivity of dairy cows. In study 2, 30 transition dairy cows were treated several times orally before parturition either with saline (controls) or with increasing doses of LPS from E. coli 0111:B4 and a flat dose of LTA from Bacillus subtilis. Results from this trial demonstrated that the treatment increased salivary and vaginal mucus total IgA, modulated selected plasma metabolites, decreased acute phase proteins and pro-inflammatory cytokines as well as lowered the incidence of lameness, metritis, and retained placenta. Results of these studies are encouraging and warrant further research.
