​​​​​​​Prominin-1 or otherwise known as CD133 is a glycoprotein that is present in humans and mice. Since the first description of prominin in 1997, in mouse neuroepithelial cells and in human hematopoietic stem cells as AC133 antigen, this molecule has aroused a large interest especially, as a stem cell marker, that gave rise to an ever growing body of publications and more recently its expression in cancer stem cells. Controversies as to its role as a cancer stem and its detection in different models, as well as its use as a prognostic marker have emerged. Yet, beyond its use as a stem cell and cancer stem cell marker, prominin-1/CD133 displays unique biological features and appears of importance in other processes like for example in retinal biogenesis. Indeed, this five-transmembrane plasma membrane glycoprotein, which marks membrane protrusions is associated with several essential processes like cell polarity, asymmetric cell division and membrane remodeling. We propose to review current knowledge about this intriguing molecule and present pertinent information to determine the biological role of prominins and assess their importance in medicine and cancer research. Pluripotent cells have the potential to create any cell type, and therefore represent a source of healthy cells by which to replace diseased cells in regenerative therapies. Thus, understanding how pluripotent cells are established and regulated is a major milestone in developing safe and effective regenerative therapies. In the mammalian embryo pluripotent cells emerge through two rounds of cell fate decisions that in addition to establishing pluripotent cells, establish two differentiated cell lineages. The molecules and signaling pathways that regulate these cell fate decisions represent key players in the establishment and regulation of pluripotent cells. In this thesis, I use the mouse embryo as a system in which to gain insight into how pluripotency is established and regulated. Cdx2, Oct4 and Sox2 are transcription factors that are important regulators of pluripotency in vitro, but their role in regulating the cell fate decisions that establish pluripotency in vivo is less clear. By using a loss-of-function approach, I define the roles of each of these factors in regulating the cell fate decisions that establish pluripotency in the early mouse embryo. First, by eliminating maternal and zygotic Cdx2 I demonstrate that Cdx2 is not a maternal determinant for pluripotency or differentiation during early embryo cell fate decisions. Second, I eliminate both maternal and zygotic Oct4 and demonstrate that the initial specification of pluripotent and non-pluripotent lineages in the early mouse embryo is independent of Oct4, and that instead, Oct4 cell-autonomously regulates both formation of pluripotent cells, and differentiation of a non-pluripotent lineage, the primitive endoderm. Third, I demonstrate that in contrast to Oct4, Sox2 promotes differentiation of primitive endoderm non-cell autonomously. These results support the model that pluripotency is acquired through these early embryonic cell fate decisions, rather than being determined during early embryogenesis. Furthermore, analysis of the roles of Oct4 and Sox2 in these early embryonic cell fate decisions highlights distinct roles for these factors in establishing pluripotency and suggests that although in vitro Oct4 and Sox2 appear to have overlapping roles, these master regulators of pluripotency have distinct cell type specific roles in regulating the cell fate decisions that establish pluripotency in the early mouse embryo. Taken together, the research presented in this thesis greatly increases our understanding of the regulation of the cell fate decisions that establish pluripotency in the embryo and provides important insights into the biology of key regulators of pluripotency in vitro and in vivo.

Studies on stem cells have been attracting intense scientific and p- lic attention, not only because of controversies surrounding the use of embryonic stem cells but also because of very provocative data that have been emerging on adult stem cells. Much of the public attention and debate has been focused on the possibility that adult stem cells may be used as a substitute for human embryonic stem cells or as a justification for stopping work on them. This has somewhat dim- ished attention on very heated scientific debates that take us to the very heart of how the concept of stem cells is perceived. To this author, the latter debates have not been unlike certain philosophical debates of the last century. Since the seminal studies of Till and McCulloch in the 1960s, the popular paradigm on adult stem cells has been that lineage-restricted stem cells are derived from pluripotent stem cells very early during development. To many, and consistent with much data, the restriction to particular lineages was considered absolute. In other words, there was a sense of determinism in the stem quality of particular stem cells: once they were allocated, they were programmed to specific roles in a given tissue. Furthermore, some adult tissues were considered devoid of detectable stem cell presence or activity.
