We have therefore reproduced some of the key articles that elegantly document these dramatic stages of development of the field in an inclusive appendix to the book, for the benefit of readers keen to investigate the history of how the field of stem cell biology has evolved. However, several roadblocks, such as safety issues and low efficiency of pluripotent stem cell (PSC) line derivation need to be resolved before their clinical application. This thesis focuses on these two areas, so as to find methods to overcome the limitation. It covers deriving embryonic stem cells (ESCs) from several different species and reports an efficient system to generate induced pluripotent stem cells (iPSCs), and the first iPSC mice in the world. The results in this thesis confirm that somatic cells can be fully reprogrammed with the four Yamanaka factors. In addition, we have found that the Dlk1-Dio3 region can be a potential molecular marker to distinguish the fully reprogrammed iPSCs from partially reprogrammed ones. All of these results will help improve the safety of PSCs in the clinical applications and increase the current low induction efficiency of their production.

Induced pluripotent stem cells (iPSCs) are a type of pluripotent stem cell artificially derived from non-pluripotent cell, typically an adult cell with regeneration capability. Active Motif, a biotechnology company that specializes in supplying epigenetic resources and high quality antibodies involved in the regulation and signaling in stem cell biology is looking to extend their product offerings by establishing a stem cell core facility to advance the development of stem cells related products. This project was aimed at establishing a stem cell core facility and validating CDy1 a pluripotent stem cell dye currently sold by Active Motif. This project expands on recent on findings on the use of CDy1 dye for stem cell sorting. Three different stem cell lines, human embryonic stem cells (hESCs), mouse embryonic stem cells (mESCs) and induced pluripotent stem cells (iPSCs) as well as a new batch of CDy1 dye was used for this project. A CDy1 dye that was licensed from a collaborator was used as a control for CDy1 dye produced by Chromeon to test the new batch's biological equivalency. The results indicate that Chromeon dye is biologically equivalent to the licensed dye and is comparable to live cells alkaline phosphatase staining and fixed cells alkaline phosphatase staining. Based on Immunocytochemistry and Flow cytometry analysis data of CDy1 co-stained with various pluripotency markers, stem cell colonies expressing CDy1 also expressed high levels of pluripotency markers. However the result is inconsistent in iPSC culture as the percentage of cells that expressed both CDy1 and various pluripotency markers were lower than expected. Future work would need to be done to optimize the condition for the iPSCs. The use of CDy 1 dye for stem cell isolation and characterization is an invaluable tool for stem cell research.

The induced pluripotent stem cells (iPSCs) have an intrinsic capability for indefinite self-renewal and large-scale expansion and can differentiate into all types of cells. Here, we tested the potential of iPSCs from dental pulp stem cells (DPSCs) to differentiate into functional odontoblasts. DPSCs were reprogrammed into iPSCs via electroporation of reprogramming factors OCT-4, SOX2, KLF4, LIN28, and L-MYC. The iPSCs presented overexpression of the reprogramming genes and high protein expressions of alkaline phosphatase, OCT4, and TRA-1-60 in vitro and generated tissues from 3 germ layers in vivo. Dentin discs with poly-L-lactic acid scaffolds containing iPSCs were implanted subcutaneously into immunodeficient mice. After 28 d from implantation, the iPSCs generated a pulp-like tissue with the presence of tubular dentin in vivo. The differentiation potential after long-term expansion was assessed in vitro. iPSCs and DPSCs of passages 4 and 14 were treated with either odontogenic medium or extract of bioactive cement for 28 d. Regardless of the passage tested, iPSCs expressed putative markers of odontoblastic differentiation and kept the same mineralization potential, while DPSC P14 failed to do the same. Analysis of these data collectively demonstrates that human iPSCs can be a source to derive human odontoblasts for dental pulp research and test bioactivity of materials.
