Different aspects of engineered viral resistance were analyzed, and transfer of this technology accomplished. Papaya plants transformed with sense/translatable coat protein (CP) genes of Papaya ringspot virus (PRSV) isolated from Venezuela and sense/untranslatable from a Hawaiian isolate were analyzed. In general, translatability did not affect the level and range of resistance, but sequence similarity between the transgene and the challenging virus did. Some isolates with high sequence similarity to the transgene, however, overcame resistance, which points to the importance of virulence determinants--probably suppressors of gene silencing--as future targets for improving engineered viral resistance. Additionally, a genomic insertion of the commercial transgenic 'Rainbow' papaya was cloned and sequenced confirming that it harbors a single CP insertion. To further analyze the impact that sequence similarity has on transgenic viral resistance a diverse array of synthetic genes derived from the nucleocapsid gene of Tomato spotted wilt virus (TSB were designed and used to transform Nicotiana benthamiana plants. The larger the number of nucleotide changes, the more detrimental effect in transgenic viral resistance, especially if changes are scattered along the synthetic gene. These findings allowed us to design and synthesize a short gene that conferred resistance to two tospoviruses that are distantly related at the level of genomic sequence. Our data extends previous observations on the feasibility of obtaining multiple virus resistance. It also shows, for the first time, that synthetic, short genes can be created to simultaneously target different virus for degradation, and endogenous genes for inactivation. The use of this approach to target for inactivation other viruses and/or endogenous genes is addressed.
