Melanoma is the deadliest form of skin cancer and it is among the cancers with the most rapidly increasing rates in the US and other western countries. Patients diagnosed and treated in the early stages have a high rate (99%) of five year survival. Once melanoma tumor metastasizes, five year survival rate dramatically drops to 20%. Notch is one of the simplest signaling pathways that is required for almost every cell type in the body. It plays critical role in several cell and developmental processes such as cell fate decision, differentiation and stem cell maintenance. Notch signaling has been implicated in melanoma tumor initiation, progression and metastasis. Preclinical and clinical studies targeting Notch signaling for treatment of melanoma have produced conflicting results. This prompted us to reevaluate the role of Notch signaling in melanoma tumor formation, growth, progression and drug resistance. Our results suggested that Upregulation of Notch signaling induces cell senescence in normal human melanocytes in culture. Knockdown of Notch1 or Notch2 in BRAFV600E/Pten null mice tends to accelerate tumor formation and growth. We also show that neither Notch1 nor Notch2 deletion can substitute for the tumor suppressor function of Pten in BRAFV600E-induced melanomagenesis. Our data showed that Notch signaling is dispensable for melanoma cell survival and that Notch signaling inhibition only affects the clonogenic ability of some primary melanoma cells in a stage non-specific manner. We showed that Notch signaling is downregulated in BRAFV600E MAPKi resistant cells and that its upregulation by NICD overexpression activates apoptosis in the MAPKi-resistant cells, but not the MAPKi-sensitive cells. Our data suggested that NICD activates apoptosis in the resistant cells in part by downregulating EDN1 through c-JUN. We evaluated the prognostic value of Notch signaling in primary melanoma patients and found that expression of NOTCH1, NICD protein or Notch effector HES1 are not reliable predictors of aggressiveness of primary cutaneous melanoma.
