Book Description :

Beginning from centuries of anecdotal descriptions of cell death, such as those on the development of the midwife toad in 1842 by Carl Vogt, to modern-day investigations of cell death as a biological discipline, it has become accepted that cell death in multicellular organisms is a normal part of life. This book provides a comprehensive view of cell death, from its mechanisms of initiation and execution, to its implication in human disease and therapy. Physiological cell death plays critical roles in almost all aspects of biology, and the book details its roles in lymphocyte homeostasis, neuronal function, metabolism, and the DNA damage response. When physiological cell death goes awry, diseases can arise, and cancer is presented as a central paradigm for the consequences of derangements in the interplay between cell survival and cell death. At the same time, the potential promise of targeted therapies aimed at interdicting cell death machineries are also discussed extensively. The molecular mechanisms that underlie apoptotic cell death are illustrated from the perspectives of both the intrinsic, mitochondrial apoptotic pathway and the extrinsic, death receptor pathway. Key players in these pathways, such as the Bcl2 family proteins, cytochrome c, Apaf-1, caspases, death receptor adapter proteins, and inhibitor of apoptosis proteins, are presented from both functional and structural angles. Until only a few years ago, programmed cell death has been considered essentially synonymous with apoptosis. However, we now know that programmed cell death can also take other forms such as necrosis or necroptosis, and to this end, the mechanisms that underlie programmed necrosis in development and host defense are illustrated. The past twenty plus years have seen an incredible growth of research in cell death, with one breakthrough after another, and the legacy still goes on with constant new surprises and findings. Long live cell death!

Book Description :

Protein Modificomics: From Modifications to Clinical Perspectives comprehensively deals with all of the most recent aspects of post-translational modification (PTM) of proteins, including discussions on diseases involving PTMs, such as Alzheimer's, Huntington's, X-linked spinal muscular atrophy-2, aneurysmal bone cyst, angelman syndrome and OFC10. The book also discusses the role PTMs play in plant physiology and the production of medicinally important primary and secondary metabolites. The understanding of PTMs in plants helps us enhance the production of these metabolites without greatly altering the genome, providing robust eukaryotic systems for the production and isolation of desired products without considerable downstream and isolation processes. Provides thorough insights into the post translational modifications (PTMs) of proteins in both the plant and animal kingdom Presents diagrammatic representations of various protein modification and estimation mechanisms in four-color Includes coverage of diseases involving post translational modifications

Book Description :

Translational medicine has opened the gateway to the era of personalized medicine. No longer a one-size-fits-all approach, the treatment of cancer is now based on an understanding of underlying biologic mechanisms and is increasingly being tailored to the molecular specificity of a tumor. Translational Oncology: Targeted Therapy in Translational Cancer Research provides a comprehensive overview of the pertinent molecular discoveries in the cancer field and explains the clinical ramifications and utility of these on targeted cancer therapies. Beginning with a chapter introducing the “Bench to Bedside and Back” paradigm, an overview is presented of the exponential growth in the field of translational medicine, including historical perspectives and recent progress. Following the introduction, subsequent chapters review the progress of targeted therapy within three major categories: hematological malignancies, solid tumors, and hereditary cancers. Each of these categories comprises a single a chapter with subsections that discuss specific cancer types. Following this is a chapter on targeted and functional imaging that reviews progress in the different modalities of anatomical and molecular imaging used to complement targeted therapy. The next chapter discusses the need to combine individual agents with existing therapies or in novel combinations in order to overcome drug resistance. Following this is a chapter presenting the challenges in drug development and the selection of targets for cancer therapeutics. The final chapter offers perspectives on personalized medicine and presents future challenges in the investigation of optimal combinations and the identification of biomarkers that can provide both predictive and prognostic information for tailoring targeted therapies to individual patients.

Book Description :

This volume explores the mechanisms of resistance to targeted therapeutics. The focus is on the cancer cell signaling network, although other mechanisms of resistance including target mutation, and new areas of study such as cancer stem cells are included. Targeted Therapies: Mechanisms of Resistance highlights examples of changes in the signaling network in response to inhibition of a signaling event and underscores the importance in having a mechanistic understanding of the signaling network in cancer for developing effective targeted cancer therapies. Moreover, cutting edge tools to analyze the cell signaling network will be discussed. This includes the leading edge of techniques as well as computational biology and systems theory. This volume provides the reader with both an overview as well as a detailed perspective on the mechanisms of resistance to targeted therapeutics and will be of great value to the oncologist, the physician-scientist treating patients and the translational scientist working on any aspect of targeted therapeutics.

Book Description :

Recent advances in the understanding of the biological basis of pediatric soft-tissue and bone tumors, especially owing to the advent of “omics” technologies, have led to an exponential increase in the current knowledge on the genetic and cellular patho-mechanisms that drive these diseases. This offers the unprecedented opportunity to develop and implement targeted therapies such as monoclonal antibodies, small molecules, oncolytic viruses, and immunotherapies in standard and/or personalized treatment regimens. However, to date only a few examples document a successful translation of discoveries from the bench to the bedside. Recent international expert congresses further emphasize the urgent need for a more rapid and especially more successful translational process. Hence, we strongly believe that a Frontiers Research Topic aiming at this aspect would fit just in time and be relevant for a broad readership. This Frontiers Research Topic intended to provide a platform for active and interdisciplinary discussion, to summarize current state-of-the-art knowledge on all basic research and translational aspects in pediatric soft-tissue and bone tumors, and to offer new perspectives on how to further promote and accelerate the translational process. It comprises high-quality original articles and timely reviews.

Book Description :

There have been tremendous advances in our understanding of molecular and tumor biology during the past few years. In the field of cancer therapeutics, it is expected that cytotoxic drug approaches will be gradually replaced with treatments based on biological targeted approaches. Hopefully these new targeted therapies will significantly increase efficacy and lack the devastating and troublesome side effects elicited by cytotoxic chemotherapy. This volume is the first book to cover the general topic of targeted cancer therapy. It presents a range of targets such as tumor angiogenesis, cell cycle control and cell signalling, COX-2, apoptosis/cell survival, invasion and metastasis and approaches like kinase inhibitors, antisense, and antibody-based therapeutics. The emphasis is on preclinical development, including target validation, development of biomarkers, strategies for combination approaches, and development of resistance. The particular challenges involved in translating these data to clinical application are discussed. This volume should be of broad general interest to researchers and clinicians involved in cancer therapy as well as other scientists interested in current strategies for cancer treatment.
