Book Description :

Chelation Therapy in the Treatment of Metal Intoxication presents a practical guide to the use of chelation therapy, from its basic chemistry, to available chelating antidotes, and the application of chelating agents. Several metals have long been known to be toxic to humans, and continue to pose great difficulty to treat. These challenges pose particular problems in industrial settings, with lead smelting known to be associated with hemopoietic alterations and paralyses, and the inhalation of mercury vapor in mercury mining being extremely detrimental to the central nervous system. Clinical experience has demonstrated that acute and chronic human intoxications with a range of metals can be treated efficiently by administration of chelating agents. Chelation Therapy in the Treatment of Metal Intoxication describes the chemical and biological principles of chelation in the treatment of these toxic metal compounds, including new chelators such as meso-2,3-dimercaptosuccinic acid (DMSA) and D,L-2,3-dimercapto-1-propanesulfonic acid (DMPS). Presents all the current findings on the potential for chelation as a therapy for metal intoxication Presents practical guidelines for selecting the most appropriate chelating agent Includes coverage on radionuclide exposure and metal storage diseases Describes the chemical and biological principles of chelation in the treatment of toxic metal compounds

Book Description :

This is an easy-to-read manual about chelation therapy. It contains specific protocols for the treatment of chronic metal overexposure, all of which are evidence-based, aiding the inexperienced and the experienced therapist in selecting the optimal chelating agent for the metal(s) in question. Expert chemists and chelation therapists have contributed invaluable knowledge and experience to this book. Most importantly, this is not a book promoting personal views or preferences. In fact, the information presents and explains different approaches, and allows the reader to draw his own conclusion. All the protocols listed have been modified for the treatment of chronic metal intoxication, included are oral and intravenous treatment plans. The authors explain in simple terms how a chelating agent ́s bioavailability affects metal binding and how various protocols such as the Cutler Protocol affect the detoxification process, why DMPS binds mercury and which chelators are useful for the detoxification of certain organ system. Also discussed is the importance and effect of treatment pauses. Diagnostic tests are discussed, and which are useful under certain conditions. Shown are tables comparing the average metal binding ability of the various EDTAs, DMPS, DMSA and other chelating agents. Detailed information explains which chelator binds which metal and why, all of which aids the therapist in finding optimal treatment schedules. Novel chelating agents are presented such as MIADMSA, a new oral chelator specifically designed for arsenic intoxication, or how natural Pectin may be used an alternative for gadolinium intoxication, which metals are bound by lipoic acid and why a slightly alkaline environment supports metal binding, even preventing the often-feared metal redistribution. All in all, this evidence-based chelation "cookbook" should be in medical libraries and on the bookshelf of every physician treating environmental disease.

Such development, and the evolution of improved strategies of Iron Chelating Therapy require better understanding of the pathophysiology of iron toxicity and the mechanism of action of iron chelating drugs. The timeliness of the present volume is underlined by several significant developments in recent years. New insights have been gained into the molecular basis of aberrant iron handling in hereditary disorders and the pathophysiology of iron overload. This volume highlights the impact of long term Iron Celating Therapy using deferoxamine or the new, but controversial oral iron chelator deferiprone based on experience gained by multicenter trails, with special emphasis on survival, morbidity and drug toxicity; it reviews the development of the new and improved orally effective chelators suitable for clinical use in the near future and examines novel strategies of iron chelating treatment for the control of cell proliferation in malignant disease or malaria.

Book Description :

Over 400 years ago, Swiss alchemist and physician Paracelsus (1493-1541) cited: "All substances are poisons; there is none that is not a poison. The right dose differentiates a poison from a remedy." This is often condensed to: "The dose makes the poison." So, why are we overtly anxious about intoxications? In fact, poisons became a global problem with the industrial revolution. Pesticides, asbestos, occupational chemicals, air pollution, and heavy metal toxicity maintain high priority worldwide, especially in developing countries. Children between 0 and 5 years old are the most vulnerable to both acute and chronic poisonings, while older adults suffer from the chronic effects of chemicals. This book aims to raise awareness about the challenges of poisons, to help clinicians understand current issues in toxicology.

Book Description :

Focusing on the practical use of N-Acetyl-Cysteine (NAC) in medicine, this book provides a comprehensive review of the basic biological and clinical studies documenting its benefits in treating medical disease. NAC is perhaps best known as an antidote for acetaminophen, but its therapeutic effect in a wide range of medical diseases has recently been realized. In addition to its well recognized use in radiological contrast prophylaxis for renal disease and pulmonary disorders, studies have suggested significant promise in psychiatric and neurological disorders such as addiction, Alzheimer’s disease, ataxia, autism, bipolar disorder, depression, epilepsy, neuropathy, obsessive-compulsive disorder, schizophrenia, traumatic brain injury and trichotillomania in addition to promising studies in audiology, cardiology, exercise physiology, gastroenterology, hematology, infectious disease, infertility and ophthalmology. Given the promising studies for a wide range of medical conditions, coupled with a excellent safety profile, the potential for NAC in the treatment of human disease appears considerable. Dr Leonore A Herzenberg from Stanford University, a pioneer of redox physiology and the use of NAC, provides a succinct history of the development of the therapeutic use of NAC for medical disease. This is followed by a series of basic science chapters outlining the role of NAC in important physiological processes, including modulation of dopamine and glutamate neurotransmitter systems, redox and mitochondrial metabolism, apoptosis and inflammation. The last section of the book is dedicated to clinically oriented chapters that comprehensively review the literature on medical disorders in which NAC has been found to be effective, including toxicity and cardiovascular, gastrointestinal, neurological, psychiatric, pulmonary and renal disorders. Each chapter reviews the theoretical biological mechanisms of NAC for the specific diseases reviewed, rates the clinical studies using a standardized criteria in order to provide an objective level of evidence and grade of recommendation for the use of NAC for specific medical conditions and outlines the ongoing clinical trials examining NAC for the treatment of specific diseases. Final chapters review the clinical evidence verifying that specific theoretical biological mechanisms are actually being targeted by NAC in medical disease. Studies on the pharmacology, formulation and potential adverse effects of NAC are also reviewed. A final chapter synthesizes the clinical studies to suggest that the effectiveness of NAC may signal a new basic physiological disorder, glutathione deficiency, which may be an important pathophysiological mechanism of many diseases.

Book Description :

Mercury is widespread in our environment. Methylmercury, one organic form of mercury, can accumulate up the aquatic food chain and lead to high concentrations in predatory fish. When consumed by humans, contaminated fish represent a public health risk. Combustion processes, especially coal-fired power plants, are major sources of mercury contamination in the environment. The U.S. Environmental Protection Agency (EPA) is considering regulating mercury emissions from those plants. Toxicological Effects of Methylmercury reviews the health effects of methylmercury and discusses the estimation of mercury exposure from measured biomarkers, how differences between individuals affect mercury toxicity, and appropriate statistical methods for analysis of the data and thoroughly compares the epidemiological studies available on methylmercury. Included are discussions of current mercury levels on public health and a delineation of the scientific aspects and policy decisions involved in the regulation of mercury. This report is a valuable resource for individuals interested in the public health effects and regulation of mercury. The report also provides an excellent example of the implications of decisions in the risk assessment process for a larger audience.
