Knowledge of the mechanisms and regulation of chloride, bicarbonate, and proton transport across cells and their membranes has become crucial to our understanding of many physiologic functions at both the cellular and systemic level. Cell pH and volume regulation and gastric, renal, ocular, and airway function are examples of processes that depend on the control and proper operation of anion and proton translocating mechanisms. Osmotic and metabolic functions of many plant and fungal cells also depend on these processes, and such systems have mechanisms in common with those found in animal cells. It has become evident that regulation of anion and proton transport at both cell and tissue levels participates in the physiologic homeostasis of the living state and that dysfunction in these processes can result in significant pathologic conditions. There have been significant strides in knowledge afforded by developments for ion-detecting dyes, signal channel measurements, and the application of genetic and biochemical approaches to the study of anion and proton transport systems. One possible avenue for heavy metal decontamination of the environment is phytoremediation, which is a technology based on the remarkable abilities of certain plant species to tolerate and accumulate extremely high concentrations of heavy metals. One of the best known heavy metal hyperaccumulator plant species is Thlaspi caerulescens, which is a Zn/Cd-hyperaccumulator that can accumulate and tolerate up to 30,000 ppm Zn and 10,000 ppm Cd in the shoots without exhibiting toxicity symptoms. The research described in this dissertation focuses on identifying gene(s) that may be responsible for the extreme heavy metal accumulation phenotype in Thlaspi caerulescens.
