This thesis presents various applications of graphene-based nanomaterials, especially in biomedicine. Graphene and its derivatives have gained enormous attention from scientists in all fields of study due to many unprecedented properties. The initial scientific attention was focused on the development of transparent flexible electrodes by exploiting two-dimensional graphene film’s extraordinary electrical and physical properties. Recently, given an increasing evidence of dispersed graphene-based nanomaterials’ biocompatibility, researchers have endeavored to employ these materials in other studies relevant to biomedical technologies. In this respect, the thesis provides a comprehensive review on the synthesis, toxicity, and a few of the key biomedical applications in the first chapter. The following chapter discusses the use of a graphene film as a novel catalyst to oxidatively destroy phenols, which are known to be potentially mutagenic and carcinogenic. Finally, and most importantly, the last chapter introduces the therapeutic role of graphene quantum dots, the smallest graphene-based nanomaterials, for Parkinson’s disease. The results are promising for the use of graphene quantum dots as the basis of future clinical drug candidates for neurodegenerative disorders. ​

Abstract: Layered two‐dimensional (2D) nanomaterials, such as graphene, transition metal dichalcogenides (TMDs), layered metal oxides (LMOs), black phosphorus (BP), and hexagonal boron nitride (hBN), have attracted tremendous interest recently owing to their unique structural morphologies and outstanding physicochemical properties. Consequently, these nanomaterials have been actively explored for different biological and biomedical applications, such as tissue engineering, drug delivery, bioimaging, and biosensing. As increasing efforts have been focused on identifying the potential bioapplications of layered 2D nanomaterials, one of the fundamental aspects that is of considerable interest and ought to be studied in greater depth is their biocompatibility and nanotoxicity. In addition, as the elucidation and understanding of the physicochemical properties of this new class of nanomaterials are still in their infancy, information on both in vitro and in vivo biocompatibility and nanotoxicity is still scarce and remains poorly understood. As such, there is an immediate need to explore and establish the biocompatibility and nanotoxicological profiles of these nanomaterials in order to develop and optimize them for specific bioapplications. Here, in this Focus Review, we will provide a broad overview of recent advances on the biocompatibility and nanotoxicity of layered 2D nanomaterials. First, a wide range of established and emerging layered 2D nanomaterials actively investigated for bioapplications will be introduced. Graphene and graphene oxide (GO) are two-dimensional nanomaterials with promising biomedical application potential, including drug and gene delivery, bio-imaging, and photothermal therapy. However, the nature of interactions between these materials and cells is poorly understood. Here, cell surface adhesion, subcellular localization, and size-dependent uptake are investigated in C2C12 cells using bovine serum albumin (BSA)-functionalized GO. Small BSA-GO nanosheets enter cells through clathrin-mediated endocytosis (CME), while large nanosheets enter cells through both CME and phagocytosis. Cytotoxicity is minimal. AFM-based size/thickness characterization of BSA-GO is performed in a systematic fashion, and a practical protocol for such analysis is presented. Size-dependent uptake results provide needed information about fundamental cellular interactions between cells and two-dimensional nanomaterials and will be useful in future biomedical and toxicological studies. BSA-functionalized GO is employed as a model system for quantifying surface charge distribution in biologically-relevant materials, and preliminary surface potential measurements by Kelvin probe force microscopy are presented.

Abstract : Two-dimensional (2D) graphene-like nanomaterials show wide applications in the fields of nanodevices, sensors, energy materials, catalysis, drug delivery, bioimaging, and tissue engineering. Recently, many studies have been focused on the synthesis and application of 2D transition metal dichalcogenide (TMD) nanosheets for various biomedical applications. In particular, 2D TMD nanosheets exhibit great advantages for tumor imaging and therapy compared to some traditional nanomaterials due to their high specific surface area, good biocompatibility, easy modification, and ultrahigh light and heat conversion efficiency. In this review, we summarize the recent advances in the synthesis, modification, and photo/thermo-based tumor imaging and therapy of 2D TMD nanosheets. The important studies on tumor bioimaging with TMD nanosheets, such as X-ray computed tomography, magnetic resonance imaging, and photoacoustic imaging, are demonstrated and discussed. In another section, the physical photothermal and photodynamic therapies as well as the pharmacological therapy of tumors with TMD nanosheet-based nanohybrids are introduced. It is expected that this work will be valuable for readers to understand the synthesis and modification of TMD nanosheets to design novel 2D functional nanomaterials for photo/thermo-based tumor imaging and therapy in one aspect, and in another aspect will extend the applications of TMD-based nanomaterials in materials science, analytical science, electrocatalysis, tissue engineering, and others.
