Depressive disorders affecting women during pregnancy and the postpartum period may lead to mother-infant bonding disorders and future psychological and cognitive problems for the children of affected women. Alpha amylase can be used as a surrogate for Sympathetic Nervous System (SNS) activity that is known to have a role in depressive disorders. This biomarker may be useful in early detection of perinatal depressive disorder. This primary study objective was to investigate SNS activity over the perinatal period and whether these changes are related to perinatal depression and mother-infant bonding. A further objective was to refine the alpha amylase assay in order to simplify the procedure. Fifty one women completed psychological questionnaires and provided saliva samples data throughout the pregnancy (36 weeks gestation) and postpartum periods (6 weeks postpartum). Salivary alpha amylase concentrations were measured and investigated in relation to depression and maternal bonding. Salivary alpha amylase showed a distinct diurnal pattern throughout the perinatal period. A significant positive correlation was observed between alpha amylase levels and maternal bonding. There was no significant correlation observed between alpha amylase levels and perinatal depression. There was no successful outcome regarding refining the alpha amylase method. The most interesting finding of this study was the relationship between decreased levels of salivary alpha amylase concentrations and poor mother infant bonding. Given the relatively small sample of depressed women, this finding warrants investigation in larger studies.
