Book Description :

Chirality is a fundamental, persistent, but often overlooked feature of all living organisms on the molecular level as well as on the macroscopic scale. The high degree of preference for only one of two possible mirror image forms in Nature, often called biological homochirality is a puzzling, and not yet fully understood, phenomenon. This book covers biological homochirality from an interdisciplinary approach - contributions range from synthetic chemists, theoretical topologists and physicists, from palaeontologists and biologists to space scientists and representatives of the pharmaceutical and materials industries. Topics covered include - theory of biochirality, origins of biochirality, autocatalysis with amplification of chirality, macroscopic (present) biochirality, fossil records of chiral organisms - paleochirality, extraterrestrial origin of chirality, exceptions to the rule of biological homochirality, D-amino acids, chemical transfer of chirality, PV effects, and polarised radiation chemistry.

Book Description :

Chirality is widely studied and omnipresent in biological molecules. However, how the retention of enantiomeric forms persists in many life processes without racemization is still unclear, and the molecular understanding of the stringent chiral specificity in enzymatic reactions is sparse. An overview of the influence of chirality in driving reactions within enzymatic cavities, Chirality in Biological Nanospaces: Reactions in Active Sites covers: Influences of molecular chirality on the structure of the active site and network of interactions to drive reactions with improved speed, accuracy, and efficiency The conserved features of the organization of the active site structures of enzymes The intricate interplay of electrostatic, hydrophobic, and van der Waals interactions Interactions between the active site residues and the substrate molecules Despite being time-consuming and expensive, trial-and-error is often the primary method used to develop synthetic enzymes. This book describes methods that combine crystallographic studies with electronic structure-based computational analysis. These methods may lead to future elucidation of new drugs that can target biological active sites with better efficacy and can be used to design custom-made novel biocytes with improved efficiency.

Book Description :

Chirality, or handedness, is a fundamental physical characteristic, which spans the length scales ranging from elementary particles to the chiral asymmetry of spiral galaxies. The way in which chirality in chemistry, or molecular handedness, may have emerged in a primitive terrestrial environment, and how it can be triggered, amplified, and transferred, are deeply challenging problems rooted in both fundamental scientific interests and the technological potentials for science and society. Chirality constitutes a unifying feature of the living world and is a prime driving force for molecular selection and genetic evolution in biology. In this book, we offer a selection of five distinct approaches to this problem by leading experts in the field. The selected topics range from protein chirality and its relevance to protein ageing, protein aggregation and neurodegeneration, entropy production associated with chiral symmetry breaking in closed systems, chiral oscillations in polymerization models involving higher-order oligomers, the mirror symmetry breaking in liquids and its implications for the development of homochirality in abiogenesis, the role of chirality in the chemical sciences, and some philosophical implications of chirality.

Book Description :

An integrated view of chiral drugs—from concept and synthesisto pharmaceutical properties Chirality greatly influences a drug's biological and pharmacological properties. In an effort to achieve more predictable results from chiral drugs, the Food and Drug Administration now requires that these medicines be as pure as possible, which places great demands on drug synthesis, purification, analysis, and testing. To assist researchers in acquiring the essential knowledge to meet these rigid guidelines, Chiral Drugs focuses on three vital chiral technologies—asymmetric synthesis, biocatalytic process, and chiral resolution—to offer details on the basic concepts, key developments, and recent trends in chiral drug discovery, along with: The history of chiral drugs development and industrial applications of chiral technologies A section listing twenty-five approved or advanced-trial chiral drugs that lists each drug name, chemical name and properties, a representative synthetic pathway, pharmacological characterizations, and references An interdisciplinary approach combining synthetic organic chemistry, medicinal chemistry, and pharmacology Nearly two-thirds of the drugs on today's market are chiral drugs. Reducing and eliminating their negative characteristics is an ongoing and serious challenge for the pharmaceutical industry. With its well-balanced approach to covering each important aspect of chirality, Chiral Drugs champions important strategies for tipping the medical scale in a positive direction for the production of more effective—and safer—drugs.

Book Description :

This book discusses the influence of chirality and intermolecular interactions in biomimetic (amphiphilic and peptide systems) and biological (amino acids and model biological reactions involving amino acids and peptides) systems using computational chemical methods. Generation and retention of homochirality are fundamentally important in biology. The difference in intermolecular interactions between enantiomeric and racemic pairs (homochiral and heterochiral interactions) and their interactions with the surrounding molecular structures leading to chiral discrimination, which could be responsible for homochiral preference, is also discussed. The nature of intermolecular energy surfaces as a function of molecular mutual orientations dictates the chiral discrimination in biology. The knowledge gained in the biomimetic systems is employed to understand more complex biological reactions like synthesis of amino acids by transamination reaction, synthesis of aminoacyl-tRNA in the aminoacylation process and synthesis of peptides in ribosome. The molecular recognition process in biomimetic amphiphiles and peptides are correlated with the nature of the intermolecular energy surfaces.

Book Description :

This book addresses some important open questions in this interdisciplinary field of research. In spite of its broad scope, ranging from the earliest evidence of life on earth to the search for extraterrestrial intelligence, the main focus is on chemical evolution. Once the macromolecules of life were formed, the evolution of the earliest life forms enhanced the importance of chirality. This led to the highly asymmetric environment of the macromolecules of the living cell the hallmark of life itself. The subject of chirality, in particular, is discussed in depth: the status of the weak force as the only true chiral influence is presented. A substantial number of papers review both the theoretical as well as the experimental basis of the origin of biochirality. A second broad area discussed in detail is the RNA world. Some successes of this hypothesis are highlighted; the hierarchy of previous evolutionary stages leading to the origin of life, such as the pyrophosphate world, are considered. The question is raised whether useful hints may still be inferred from molecular fossils existing in contemporary cells. Contents The Origin, Evolution, and Distribution of Life in the Universe C. Book Description :

The phenomenon of chirality occurs in many disciplines of natural sciences, such as elementary particle physics, chemistry of molecules, biopolymers and crystals, pharmacology, biology (snails, winding plants), and medicine (handedness). Review articles are widely available for most of these fields and written for the respective experts. This book gives for the first time an interdisciplinary comprehensive treatment of chirality. Ten well-known scientists describe the present state of the art in different fields in introductory review articles without going into detail. Most importantly, the book is more than an accumulation of different chapters insofar as biomolecular homochirality is traced back to the chirality of elementary particles in atomic nuclei. The authors have attempted to present chemistry for physicists and physics for chemists, etc. Any scientist who is engaged in chirality may benefit from the present survey.
