Due to the steadily growing importance of bioconjugate techniques in a wide variety of scientific applications, the development of new linkers, which facilitate specific interaction between biologically relevant molecules, has become of interest and several new linkers with unique features have been proposed and synthesized. Although there are several techniques today that are well established and commercialized for this purpose and many coupling reagents have been developed to conjugate biomolecules, there still remains a need for more specific linkers that are more suitable for certain medical applications based on the targeted active groups, reactivity and the protein-to-protein proximity that is needed for certain linkers. These synthesized new linkers are also designed such that they would provide "infinite affinity" binding. The linkers proved suitable for infinite affinity techniques utilizing single cysteine mutant single chain variable fragment (scFv) of the antibody T84.66 and its target carcinoembryonic antigen (CEA) for "infinite affinity" bioconjugation. Optimized linkers have been proposed for further consideration for bioconjugation experiments involving cysteine mutant proteins.
