More than one hundred short-term bioassays are now available for detecting the toxicity, mutagenicity, and potential carcinogenicity of chemicals. These bioassays were developed and validated with individual compounds, and their principal application was perceived to be in evaluating the health hazard of such materials. However, man is rarely exposed to single chemicals; his exposure to hazardous chemicals is more commonly a multifactorial phenomenon. Although chemical analysis can be used to detect known hazardous compounds, it would be a staggering and expensive task to analyze large numbers of samples for all known or suspected hazardous constituents. Furthermore, the biological activity of a complex mixture cannot be reliably predicted from knowledge of its components. On the other hand, bioassays alone cannot tell us which components of complex mixtures are responsible for the biological activity detected. Thus, cost effectiveness and technical feasibility dictate stepwise and perhaps iterative application -of both chemical and biological methods in evaluating the health effects of complex environmental mixtures. Through the coupling of reliable biological detection systems with methods of chemical fractionation and analysis, it is frequently possible to isolate the individual chemical species that show biological activity. Initially, complex mixtures may be separated and bioassayed in carefully defined chemical fractions. The results of such short-term screening bioassays then may be used td guide the course of further fractionation and to determine the need for more stringent and comprehensive biological testing.

Many types of tests can be considered as forms of bioassay. The broad range of substrate types, chemical composition, geographical locations, and equipment used in dredging adds to the complexity of formulating a single bioassay procedure applicable to all dredging programs. A discussion of the bioassay principles and techniques in the literature are presented to provide a background for an understanding of aquatic bioassay testing. Aquatic bioassays, which were developed from pharmacological drug testing techniques, often encounter complex toxicity problems. Two or more toxic compounds may be acting together or the toxicant concentration may fluctuate. The techniques used in aquatic testing may vary depending on the type of toxicant tested and the environmental parameters, such as temperature or dissolved oxygen, that need be controlled. Toxicant dosing equipment varies with the type of test species, source of water, and type of toxicant in question. Static bioassays are generally performed in glass jars or commercial aquarium tanks. Continuous-flow equipment consists of a reservoir of toxicant water which is metered into the test vessel. Extensive elaboration of the equipment has led to multiple toxicant delivery systems capable of providing a range of concentrations. The two most widely used delivery systems are the serial diluter and proportional diluter.

Sediment from the Illinois Waterway navigation channel is hydraulically dredged by the US Army Engineer District, Rock Island, and placed in the nearshore environment via pipeline. Water returning to the river can have a high-suspended solids load approaching fluid mud consistency. There is a concern that this return water may exceed the State of Illinois water quality standards for ammonia and have adverse effects on aquatic life. To address these concerns, composite sediment samples and site water collected from selected sites in the Illinois Waterway were evaluated in toxicity tests. Acute (48-hr) toxicity tests were conducted with two species, Pimephales promelas (the fathead minnow) and Daphnia magna (a freshwater cladoceran). A chronic (21-day) toxicity test was also conducted using Daphnia magna. Animals were exposed separately to different concentrations of filtered and unfiltered elutriates prepared from Acute, Cadmium, Daphnia magna, Pimephales promela, Ammonia, Chronic, Elutriate, Sediment, Bioassay, Cladoceran, Fathead minnow. Illinois Waterway edged material. Total ammonia concentrations were measured in all tests and the un- ionized fraction was calculated by adjusting for temperature and pH. Tests were conducted at the US Army Engineer Waterways Experiment Station, Vicksburg, MS. In addition, as part of an interlaboratory effort, a 48-hr acute toxicity test with Pimephales pomelas fry was conducted concurrently by the Hygienic Laboratory of the University of Iowa, Des Moines, IA.
