Book Description :

In this volume, more than 50 leading international experts review the latest scientific and clinical observations on inherited ataxias. The book demonstrates how molecular genetic studies, as well as recent physiological, neurochemical, and clinical data, have generated new concepts on the nosology of these disorders. Close attention is given to the important practical applications of these new findings - in diagnosis, prognosis, and genetic counseling, in development of tests for prenatal diagnosis and carrier detection, and in the search for more effective therapies. The opening chapter identifies the clinical features that distinguish the various inherited ataxic syndromes and presents a classification based on etiology, mode of inheritance, age of onset, and associated clinical features. A major portion of the book focuses on current clinical and molecular genetic studies of different forms of inherited ataxia. Coverage includes a molecular analysis of the Friedreich's ataxia locus and extensive studies on autosomal recessive spastic ataxia of Charlevoix-Saguenay, ataxia telangiectasia, dominantly inherited spinocerebellar ataxias, Machado-Joseph disease, and inherited prion diseases. The contributors provide detailed information on the various clinical phenotypes of each form of inherited ataxia and thoroughly explain the use of linkage analysis and other molecular genetic techniques to localize and isolate the genes responsible for these diseases. The book also reviews the most significant research findings on neurotransmitters in the cerebellum, on the phosphoinositide second messenger system in cerebellar degenerative disorders, and on oligodendrocyte-associated and myelin-associated inhibitors of neurite growth in the adult nervous system. The contributors assess recent progress in developing drugs for treatment of ataxias and other cerebellar movement disorders and identify new targets for pharmacological intervention. Experimental therapeutic observations on cerebellar grafting in heredodegenerative ataxia are also presented. This volume is an invaluable reference for clinicians treating patients with ataxias or counseling families at risk for inherited neurological diseases. It is also a rich source of ideas for molecular geneticists and for neuroscientists investigating disorders of the cerebellum.

Book Description :

"This book on cerebellar disorders begins with our current understanding of the anatomy and physiology of the cerebellum, and then dives into the diagnosis and treatment of the many different disorders. As virtually all pathological processes can affect the cerebellum, the book actually covers much of neurology. Dr. Manto has been devoted to the study of the cerebellum in his career. His contributions are numerous in many areas, and his enthusiasm led him to found and edit the journal, The Cerebellum, which has rapidly become quite distinguished with an impact factor approaching 4. When dealing with the different disorders, the book approaches the patient in multiple ways. What is the differential diagnosis by age or by pathologic entity? What is the differential diagnosis for recessive, dominant, or X-linked disorders? What laboratory tests should be done to clarify the situation? What scales can be used to quantify the disorder? And then, what can be done for treatment? There is no reason to take a dimview of treatment when the possibilities are taken as a whole for all the ataxias"--Provided by publisher.

Book Description :

Mitochondrial cytopathies are mutations in the inherited maternal mitochondrial genome, or the nuclear DNA-mutation. Mitochondrial respiratory chain disorders (RCD) are a group of genetically and clinically heterogeneous diseases, due to the fact that protein components of the respiratory chain are encoded by both mitochondrial and nuclear genomes and are essential in all cells. In addition, the biogenesis, structure and function of mitochondria, including DNA replication, transcription, and translation, all require nuclear encoded genes. Since mitochondria are present in every cell, every tissue, mitochondrial disorder usually affects multiple organs.

Book Description :

Understanding the Cerebellar Disorders in Large Context: Dysmetria of Thought, Cerebellar Cognitive Affective Syndrome, and Ataxia. A Concise Review. Many diseases involve the cerebellum and produce ataxia, which is characterized by incoordination of: · Balance, · Gait, · Extremity, · Eye movements, and · Dysarthria. Cerebellar lesions do not always manifest with ataxic motor syndromes, however. The cerebellar cognitive affective syndrome (CCAS) includes impairments in executive, visual-spatial, and linguistic abilities, with affective disturbance ranging from emotional blunting and depression, to disinhibition and psychotic features. The cognitive and psychiatric components of the CCAS, together with the ataxic motor disability of cerebellar disorders, are conceptualized within the dysmetria of thought hypothesis. Damage to the cerebellar component of the distributed neural circuit sub-serving sensorimotor, cognitive, and emotional processing disrupts the universal cerebellar transform, leading to the universal cerebellar impairment affecting the lesioned domain The universal cerebellar impairment manifests as ataxia when the sensorimotor cerebellum is involved and as the CCAS when pathology is in the lateral hemisphere of the posterior cerebellum (involved in cognitive processing) or in the vermis (limbic cerebellum). Cognitive and emotional disorders may accompany cerebellar diseases or be their principal clinical presentation, and this has significance for the diagnosis and management of patients with cerebellar dysfunction. An effort has been made to survey the current understanding of the cerebellar role in cognitive and psychiatric disorders. To place the consideration of the cerebellum in a larger context, however, it is attempted to first begin with a description of the cerebellar motor syndrome and a survey of the more common cerebellar diseases along with relevant ILLUSTRATIONS for better understanding. …Dr. H. Book Description :

The stimulus for writing this book arose from the author's perception of a lack of available texts which adequately integrate the subjects of neuroanatomy and functional neurology with the practice of speech language pathology. This perception was gained from almost two decades of teaching in the areas of neuroanatomy and acquired neuro logical speech-language disorders to speech pathology students initially at the South Australian College of Advanced Education and, for the past five years, at the University of Queensland. Although a plethora of excellent texts devoted specifically to each of the subjects of neuroanatomy, neurology and aphasiology have been published, few have attemped to integrate these individual subject areas in such a way as to provide a more clear understanding of the neurological bases of clinically recognized forms of aphasia and motor speech disorders. In writing this text, I have attempted to provide a better balance between neuroanatomy-neurology and. speech-language pathology. Relevant areas of neuroanatomy and neurology are introduced and discussed in the context of specific speech and language disorders. In this way, I have aimed at providing a better link between the relevant neuroanatomical and neurological knowledge on the one hand, and specific neurologically based communication disorders on the other, in order to enhance the reader's understanding of the origins, course and prognosis of these disorders. Of course the writing of any book requires the support and encouragement of other people. This text was no exception.
