Book Description :

Antimalarial Agents: Design and Mechanism of Action seeks to support medicinal chemists in their work towards antimalarial solution, providing practical guidance on current developments and highlighting promising leads for the future. Malaria is a deadly disease which threatens half of the world’s population. Advances over the last decade have seen vast improvements in the effectiveness of both preventative measures and treatments, but the rapid adaptability of the disease means that the ongoing search for improved and novel antimalarial drugs is essential. Beginning with a focus on biological aspects of malaria, this book highlights the lifecycle of the parasite responsible for malaria, the problem of resistance, genetic mapping of the parasite’s genome, established drug targets, and potential drug targets for the future. The book also includes detailed study of the medicinal chemistry of antimalarial agents and a focus on the design of antimalarial drugs. Drawing on the knowledge of its expert authors and coupling historic research with current findings to provide a full picture of both past and current milestones, Antimalarial Agents: Design and Mechanism of Action is a comprehensive yet accessible guide for all those involved in the design, development and administration of antimalarial drugs, including academic researchers, medicinal chemists, malaria researchers and pharmaceutical scientists. Consolidates both past and current developments in the discovery and design of antimalarial drugs Presents content in a style that is both thorough and engaging, providing a supportive and guiding reference to students and researchers from interdisciplinary backgrounds Highlights drug targets currently considered to be the most promising for future therapies, and the classes of compounds that are currently being studied and perfected

Book Description :

Medicinal Chemistry, Volume 12: Antimalarial Agents: Chemistry and Pharmacology presents the essentials of both biology and chemistry pertinent to the chemotherapy of malaria. This book discusses the nature of the disease, the physiology and biochemistry of the plasmodia, and the mode of action of drugs. Organized into 19 chapters, this volume begins with an overview of the most intensive efforts to develop synthetic antimalarial drugs. This text then examines how drugs are evaluated as well as the specific chemotherapy in malaria. Other chapters consider the diversity of chemical structures exhibiting antimalarial activity with emphasis on structure–activity relationships and methods of synthesis. This book discusses as well the plasmodial effects by quinine in vivo. The final chapter deals with the miscellaneous structures known to have activity against some types of plasmodial infection in animals. This book is a valuable resource for chemists and biologists involved in the development of antimalarial drugs.

Book Description :

The construction of this volume has been guided by two personal convictions. Experience in the field of experimental chemotherapy, both in the pharmaceutical industry and academia, has convinced us that recent quantum technological advances in biochemistry, molecular biology, and immunology will permit and, indeed, necessitate an increasingly greater use of rational drug development in the future than has been the custom up to now. In Part l, therefore, we asked our contributors to provide detailed reviews covering the biology of the malaria parasites and their relation with their hosts, the experimental procedures including culture techniques that are necessary to take a drug from primary screening to clinical trial, and an account of antimalarial drug resistance. Our second conviction is that many research workers are all too loath to learn from the lessons of the past. For this reason we asked the contributors to Part 2 of this volume to review very thoroughly the widely scattered but voluminous literature on those few chemical groups that have provided the antimalarial drugs in clinical use at the present time. Much can be learned from the history of their development and the problems that have arisen with them in man. Some indeed may still have much to offer if they can be deployed in better ways than they are at present. This question has been taken up by several authors.

Book Description :

Malaria has defeated previous efforts at eradication and remains a massive global public health problem despite being readily preventable and treatable. It is a devastating disease that also extracts huge economic costs from the poorest countries in endemic regions. Starting with an overview of the disease and its current political, financial and technical context, this Milestones in Drug Therapy volume describes the history, chemistry, mechanisms of action and resistance, preclinical and clinical use, pharmacokinetics and safety and tolerability of the current range of antimalarial drugs. There is particular emphasis on artemisinins and related peroxides, as these drugs have now become the frontline treatment for malaria. Next generation antimalarials, molecular markers for detecting resistance, the importance of diagnostics and disease prevention are also covered in detail.

Book Description :

This study is concerned with the preparation of: a) peroxides based on qinghaosu, b) peptide pro-drug derivatives of primaquine, and c) thymidylate synthetase inhibitors as potentially new antimalarial agents. During this study, thirty (30) target compounds were prepared and submitted for antimalarial evaluation. The thirty compounds included twenty-four (24) qinghaosu analogs, three (3) primaquine derivative and three (3) thymidylate synthetase inhibitors. Twenty-one of the qinghaosu analogs and all three of the thymidylate synthetase inhibitors were evaluated in the malaria in vitro drug screen. All but three of the qinghaosu analogs showed activity in this screen, whereas the thymidylate synthetase inhibitors were inactive. The three peptide pro-drug derivatives of primaquine were tested for radical curative activity against Plasmodium cynomologi in rhesus monkeys, All three compounds showed cures greater than that expected for the primaquine content of each pro-drug. Twenty-three target compounds were evaluated for blood schizonticidal suppressive activity against P. berghei in mice, and ten compounds were tested for causal prophylactic activity against P. berghei yoelii in mice. None of the compounds tested showed significant activity in these screens. Keywords: Malaria; Antimalarial Drugs; Heterocycles; Synthesis(Chemistry).

Book Description :

Despite over a hundred years of drug development, malaria remains one of the most devastating infectious diseases in the world, both in terms of mortality and morbidity, and its worldwide occurrence in tropical and subtropical regions. During the last few decades, chloroquine and other aminoquinolines were the front-line antimalarial agents because of their excellent therapeutic efficacy and lower cost. However, the emergence and spread of resistance of malaria parasite, especially, most deadly species, P. falciparum towards currently available drugs, especially chloroquine, has become a major health concern around the globe and therefore, development of new chemotherapeutic agents is an urgent need to fight against resistant malaria.

Book Description :

Interest in the 4-amino-1,5-naphthyridines as potential prophylactic antimalarial agents has been generated by the structural similarity of the ring system to both the 4- and 8-aminoquinolines. In synthetic studies during the program, the authors sought to expand upon the derivatives of 4-amino-1,5-naphthyridine which are currently available in an effort to secure an optimal prophylactic antimalarial drug. Accordingly, as a result of the synthetic efforts, a total of 26 naphthyridine target and 145 naphthyridine intermediates and miscellaneous precursors have been prepared and submitted for biological testing. While antimalarial testing has not yet been completed, results obtained for most of the target drugs submitted to WRAIR have disclosed no significant prophylactic activity in the lower dosage range.

Book Description :

The objectives of this program are to screen compounds against normal and drug-resistant strains of malaria and to develop procedures for use in low-temperature storage of plasmodia. Secondary screening of chemicals against normal P. berghei was continued. Test procedures were developed and utilized for screening against chloroquine-, triazine-, and DDS-resistant P. berghei in mice. A serum-free preservative was developed that maintained the viability and virulence of sensitive and drug-resistant malaria after storage at -196C for up to 225 days. This solution was also used to determine whether a splenic bank could be used as an alternative to an erythrocytic bank for the low-temperature storage of plasmodia. (Author).
